| China is a big rice growing country with huge production of rice bran as the main by-product of rice processing.Oryzanol is a mixture of ferulic acid esters with triterpene(alkene)alcohol as the main body,which is widely existed in rice bran oil.Oryzanol has a variety of physiological activities such as antioxidant,hypolipidemic,etc.As biomass materials widely used in food and medicine,functional materials and pharmaceutical and health care fields,the low solubility,poor light and heat stability limited its utilization form and the bioavailability.In this research,the electrospinning technique was utilized to fabricate the fiber membrane with the addition of inclusion compound synthesized by Oryzanol and Cyclodextrin(CD),and a series of structural and property characterization were performed to test the synthesis effect before mouse wound dressing and grape preservation tests carried out.The main research content is as follows:(1)Diphenyl carbonate(DPC)in the molten state was cross-linked with cyclodextrin by oil bath heating under a molar ratio of CD : DPC of1 : 10 as the optimal modified ratio.The optimal preparation conditions for the synthesis of the inclusion complex of oryzanol and cyclodextrin were the mass ratio of oryzanol to modified β-CD under 1 : 2,the stirring time of 16 h and the heating temperature under 25 ℃ determined by orthogonal experiments.The drug loading rate of the inclusion complex was 43.18 %and the encapsulation rate was 55.70 % under this process condition.(2)Electrospinning fiber membranes were prepared using polycaprolactone(PCL)as the substrate,and the effects of solution concentration,working voltage,plate receiving distance and injection rate on the formation of the prepared fiber membranes were investigated.The spinning solutions of polycaprolactone-chitosan(PCL-CS),polycaprolactone-oryzanol/cyclodextrin inclusion compounds(PCL-IC)and polycaprolactone-chitosan-oryzanol/cyclodextrin inclusion compounds(PCL-CS-IC)were prepared.The process parameters for the preparation of PCL fiber membrane were 16 k V,0.013 m L/min injection rate,and 15 cm plate receiving distance;the process parameters for the preparation of PCL-CS fiber membrane were 15 k V,0.014 m L/min injection rate,and 15 cm plate receiving distance;the process parameters for the preparation of PCL-IC fiber membrane were 14 k V,0.013 m L/min injection rate,and 15 cm plate receiving distance;the process parameters for the preparation of PCL-CS-IC fiber membrane were 14 k V,0.013 m L/min injection rate,and 15 cm plate receiving distance.(3)By comparing the antibacterial property,water contact angle and air permeability of the material with the control group,PCL-CS-IC fiber membrane has a relatively obvious antibacterial effect on both E.coli and S.aureus,which met the national standard antibacterial requirements;the water contact angle was 86.13°±2.65°,which was closer to the critical point between hydrophobic and hydrophilic materials with stability and facilitation of the exchange of internal and external substances;the internal fiber distribution of fiber membrane was dense,resulting in the lowest permeability with a certain shielding effect of external pollution.(4)In the rat wound dressing experiment,the rats in the PCL-CS-IC experimental group had the highest degree of skin tissue reepithelialization,the thickest stratum corneum,the highest structural integrity of the dermis and epidermis,the basic healing of the deep subcutaneous tissue,and a significant reduction in inflammatory factors in the skin tissue(P<0.05).The rat wounds were basically healed within 14 days,and by comparing the results of bacterial counts and H&E staining,it was shown that the mixture of oryzanol and chitosan was effective in reducing inflammation in the rat wounds.(5)In the grape preservation experiment,the average weight loss,decay rate,Vitamin C content and titratable acid content of grapes treated with PCL-CS-IC fiber film were 3.14 %,10 %,7.5 % and 13.56 %,respectively.The results indicated that PCL-CS-IC fiber film could effectively reduce the loss of water,Vitamin C and titratable acid of grapes during storage.It also delayed the decay process and extended the shelf life by preserving the flavor and nutritional value of grapes.In briefly,utilizing components from natural substances in nature as raw materials for pharmaceuticals,foods,etc.,which was hoped that the results of the study would provide guidance for the preparation of wound healing in pharmaceutical clinics and the development of food packaging materials. |
