Artichoke is an ancient medicinal and edible plant belonging to the genus Asteraceae.It is rich in vitamins,minerals,flavonoids,polyphenols and other nutrients,and has multiple biological functions such as liver protection,anti-cancer,and aids in digestion The content of dietary fiber(DF)in Artichoke is rich and high quality fiber.In this paper,the extraction process of Artichoke soluble dietary fiber(ASDF)was optimized by using Artichoke processing by-products as raw materials.The optimum extraction conditions of ASDF by ultrafine pulverization-microwave method were studied.The structure and functional properties of ASDF under four different extraction conditions were compared.Effects of ASDF on growth characteristics,lipid levels and liver of mice with alcoholic fatty liver;The main conclusions are as follows:(1)Using ordinary pulverization,ordinary pulverization-microwave,ultrafine pulverization,and ultrafine pulverization-microwave method to extract ASDF,it is concluded that ultrafine pulverization-microwave treatment has the highest ASDF content.The yield of ASDF extracted by microwave method increased by 9.35%;The by-products of artichoke processing were used as raw materials,and ASDF was extracted by ultrafine grinding-microwave method.Optimal process for optimizing SDF extraction.Through variance analysis,it can be seen that the quadratic polynomial regression fitted by the experiment is highly significant(P<0.001),and the SDF yield is affected by various factors in the following order:citric acid mass fraction>microwave time>microwave power>solid-liquid ratio,on the basis of single factor,the optimal process conditions of ASDF were optimized by response surface as follows:citric acid mass fraction 1%,solid-liquid ratio 1:25,microwave time 30 s,microwave power 500 W,based on this condition Model validation test,the highest yield is 19.37%.(2)The morphological structure,physical and chemical properties and functional properties of ASDF extracted by four different extraction methods were compared.The results showed that the functional properties and physical and chemical properties of ASDF extracted by ultra-micro comminution and microwave were significantly improved.From the structure analysis of ASDF,it could be concluded that the particle size of ASDF could be greatly reduced by ultra-micro comminution and microwave treatment.The particle size mainly ranged from 10 μm to 270 μm.Make its particle size smaller and more uniform distribution;The results of infrared spectrum showed that the main functional groups did not change significantly before and after extraction by different methods,and the overall peak type and position of characteristic absorption peak remained unchanged,but the intensity of ASDF absorption peak extracted by ultra-micro comminution and microwave method increased.The microstructure of ASDF extracted by superfine pulverization and microwave expanded and became loose,which resulted in the enhancement of hydration properties,significant increase of water holding capacity,oil holding capacity and expansion capacity,while the decrease of binding hydraulic capacity,and the enhancement of cation exchange capacity.Compared with ordinary pulverization,the glucose adsorption capacity is increased by 4.08 times;the antioxidant research of ASDF shows that compared with the DPPH and ABTS free radical scavenging abilities of ASDF extracted by ordinary pulverization,the ASDF extracted by ultrafine pulverization and microwave method has increased by 73.80%and 88.29%,respectively.(3)Mice with alcoholic fatty liver were fed with ASDF extracted by superfine pulverization and microwave.Under the intervention of ASDF,the liver index of mice in model group reached 0.0654,1.48 times higher than that of blank group.Meanwhile,ASDF combined with its good physical and chemical properties,The serum lipid levels in mice were effectively improved(TC,TG,LDL-C were significantly increased,HDL-C was significantly decreased).Compared with model group,SOD level of mice was increased by 53.95%,68.85%and 97.32%after low dose,medium dose and high dose ASDF intervention,respectively.Compared with the model,MDA content in positive group and ASDF dose groups decreased;Serum AST and ALT levels in low-dose,medium-dose and high-dose groups were as follows:Compared with the model group,the serum AST and ALT levels of 148.14 U/L,136.66 U/L,130.08 U/L,48.45 U/L,44.1 U/L and 40.1 UlL were decreased,and the high dose group had the most significant downward trend.The activity of GSH-Px in liver of model group was 48.00 U/mg,which was 27.59%lower than that of blank group.The activity of GSH-Px in liver of ASDF groups was 7.48%,13.35%and 26.42%higher than that of model group,respectively.After alcohol intervention,the serum LEVELS of TNF-α and IL-6 in model group were higher than those in model group.After low dose,medium dose and high dose ASDF intervention,the levels of TNF-α and IL-6 were decreased by 20.57%,25.38%,27.80%and 11.96%,17.07%,21.12%;From the above indicators,it can be concluded that high dose of ASDF has the most obvious protective effect on alcoholic fatty liver in mice.Through the observation of the HE-stained slices of the mouse liver,the hepatic steatosis,fatty vacuoles,and inflammatory cell infiltration of the mice in each dose group of ASDF were significantly improved.certain preventive protection.
