Golden pompano is one of the important coastal seafood in China,with delicious meat quality and popular among people.In the process of production and consumption,it is accompanied by a large amount of waste of by-products.In this study,with the goal of improving the high-value utilization of Golden pompano head,Golden pompano head phospholipid(GPL)was selected as the wall material and curcumin(CUR),which has excellent anti-inflammatory activity,as the core material,and Golden pompano head phospholipid-curcumin liposome(GPL-lipo)was prepared,and the comparison with soy lecithin-curcumin liposome(SPC-lipo)was conducted to evaluate the physical properties and anti-inflammatory activity at the cellular level.The details of the study are as follows:1.Preparation and optimization of Golden pompano fish head phospholipids-curcumin liposomes(GPL-lipo)Firstly,GPL-lipo was prepared by film-extrusion method,and the preparation conditions were optimized by single-factor and orthogonal experiments using the encapsulation rate of liposomes as the index,and the optimal preparation conditions were: preparation temperature of 55 ℃,preparation system PBS volume of 20 m L,CUR to cholesterol ratio of 2.5:1,and GPL to CUR ratio of 30:1.The encapsulation rate of liposomes prepared under the above process conditions The embedding rate of the liposomes could reach 87.53 % with good embedding effect.2.Physical characterization of GPL-lipo and SPC-lipoAfter that,the particle size,potential,stability and structure of GPL-lipo were evaluated by comparing with SPC-lipo.The particle size and microscopic morphology of GPL-lipo prepared in this study were similar to those of SPC-lipo,and the encapsulation effect was greater than 85%.The FTIR spectra showed that both phospholipids and CUR had no new chemical bond formation,and CUR was bound by intermolecular interaction forces.The difference is that the absolute value of the potential potential of GPL-lipo(-35.47±0.87 m V)is larger than that of SPC-lipo(-25.30±0.56 m V),indicating that the stability of GPL-lipo may be better than that of SPC-lipo.In addition,the differential scanning thermal results and storage stability tests of liposomes also showed similar results.3.Effect of GPL/SPC and CUR on NO secretion by LPS-induced RAW264.7cellsSubsequently,to investigate whether there is an interrelationship between the wall and core materials of liposomes in terms of anti-inflammatory activity,an in vitro inflammation model with LPS-induced RAW264.7 cells was established in this study to investigate the interrelationship between the effects of both on the secretion of NO in the inflammation model,using the typical inflammatory factor NO as an indicator.The experimental study showed that the co-action of phospholipids and CUR had a synergistic effect on the inhibition of NO induced by LPS in RAW264.7cells.The mechanism of action needs to be further explored,but this result still provides a new idea to explore the interrelationship between liposomal wall and core materials..4.Study on the in vitro anti-inflammatory activity of GPL-lipo and SPC-lipo and its mechanismFinally,to investigate the in vitro anti-inflammatory effects indicated by experimental studies,the effects of GPL-lipo on the secretion of NO and ROS,the expression of inflammation-related genes(TNF-α,IL-1β and IL-6)and protein levels in LPS-induced RAW264.7 cells were evaluated.Experimental studies showed that GPL-lipo significantly increased the anti-inflammatory activity of CUR.Experiments showed that GPL-lipo had a stronger anti-inflammatory effect than SPC-lipo,especially at the gene level,and the inhibitory effect of GPL-lipo on TNF-α was not only superior to that of SPC-lipo,but its effect was comparable to that of the positive control group.At the protein expression level,the inhibitory effects of both GPL-lipo and SPC-lipo were better than those of the positive control group,showing good anti-inflammatory potential,providing a technical guide for the application of GPL in liposomes and a theoretical basis for the development of functional foods.
