Radix astragali is the dry root of Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao or Astragalus membranaceus(Fisch.)Bge.,its main chemical constituents are flavonoids,saponins and polysaccharides and so on,posses effect to reinforce"Qi"(vital energy),to strengthen the superficial resistance and to promote the discharge of pus and the growth of new tissue.In this paper,the optimizing micronizing technology,the physical characteristic of micro-powder,the antioxidant activity of radix astragali micro-powder,the volatile components and its adsorption,and build the model to prodict the content of saponins by near-infrared spectrum.The main results are as follows:1.The physical characteristics of Radix astragali micro-powder was improved after grinded.The fluidity,expandability,adsorption of powder sthenghthed with the reducing of particle sizes.Dissolution rate for flavonoids of micro-powder become stronger.And the micro-powder with 400 mesh particle size had better fluidity,expandability,adsorption and higher dissolution rate for flavonoids.The angle of repose and slide were 45° and 28° partly,the swelling force was 3.9 mL g-1,the bulk denisity was 0.49 g mL-1,the water binding capacity at 30℃ was 3.02 mL g-1,the flavonoids content was 0.26%.The optimum fermentation conditions for Radix astragali mocro-powder:Select the uneaten and unmouldy roots,wash and dry,crude grind for 1min,sift out on 60 mesh,put 150g whose moisture is 6.7%to micro-grind for 20.5 min at-20℃,and the content of micro-powder with 400 mesh particle size reached 86.9%.2.The calibration model was established using the PLS(partial least squares),the study shows the multiple scattering correction pretreatment can be used to extracted spectra information to analysis the contents of saponins,the correlation coefficient was 97.84%,SEC was 1.04(standard deviation of the calibration sets).The correlation coefficient was 91.65%(standard deviation of the test sets).The forecast accuracy of this model was high,can be used to analyze saponins contents in Radix astragali micro-powder.3.30 components were identified from the volatile components of micronized Radix astragali,was 93.14%of total ion chromatogram.The most abundant components were(Z)-2-methyl-Cyclo-pentan-ol,2-butyl-2-Octenal,1-Hexadecan-ol and so on.Compared with unmicronized Radix astragali,the type of volatile components reduced and the content increased.The micro-powder of 400 particle size had sthength ABTS+·,DPPH·,·OH scavenging capacity,but it reduced compared unmicronized Radix astragali4.The appropriate adsorption conditions for Cholesterol are 400 mesh particle size,solid-liquid ratio of 0.033 g mL-1,time of 60 min,the cholesterol concentration of 1.5 mg mL-1,temperature of 25℃.And the capacity of adsorption was 15.60 mg g-1.In low concentration,The sorption was a physical surface sorption,but in high concentration it concluded surface sorption and intraparticle diffusion.The sorption can be happened spontaneous.
