| Metal complexes are compound with different spatial configurations that consist of central metal atoms or ions coordinated with organic ligands, which have the characteristics of both organic compounds and inorganic metal ions, and have good prospects in luminescent materials, catalysis, gas adsorption, etc.Among them, the nitrogen-containing heterocyclic metal complex is widely explored because of its good bioactivity, benzothiazole metal complex,as one of the branches, possess good pharmacological activities,is widely studied in antiviral, antioxidant and antitumor fields. In recent years, the mode of interaction of benzothiazole metal complexes with DNA is received more attention from scientists.DNA is a genetic material in organisms, the interaction between complex molecules and DNA will destroy the structure of DNA and affect its transcription and replication. Therefore, exploring the mode of interaction between benzothiazole metal complexes and DNA could explain the mechanism of the interaction between complex molecules and DNA,further provide a theoretical basis for designing and optimizing the structure of benzothiazole drugs. In this paper, the 2-(2-pyridyl)benzothiazole(bpt)ligand was synthesized by microwave irradiation. On this basis, five benzothiazole transition metal complexes were synthesized, named as:[Zn(bpt)(hfipbb)(H2O)](1),[Cd(bpt)(hfipbb)(H2O)](2),(Hbpt)[Mn4.5(Hhfipbb)2(hfipbb)4(bpt)3]0.5H2O(3),[Cu(bpt)(btec)0.5]·1.2H2O(4),[Zn(bpt)(NPTA)(H2O)](5).The structures of the complexes were analyzed by X-ray diffraction method. The purity of the complexes was characterized by XRD diffraction, and the thermal stability of the complexes was analyzed by thermogravimetric analysis. The interaction mode between five benzothiazole transition metal complexes and DNA molecules was studied by ultraviolet-visible spectra, fluorescence spectroscopy, circular dichroism spectroscopy and viscosity method. The experimental results showed that the five benzothiazole transition metal complexes interacted with ct-DNA in intercalation manner, which would affect the base pair stacking and right-handed helicity of ct-DNA,further damaging the double helix structure of ct-DNA,resulting in the change of its configuration, and inhibit the transcription and replication of ct-DNA.Therefore,it is hoped that the interaction between these benzothiazole metal complexes and ct-DNA in intercalation mode could provide a theoretical basis for the research and screening of antiviral, antiproliferative and other drugs. |
