| With the use of antibiotics,the problem of bacterial resistance has also emerged.The monitoring data of the national drug resistance monitoring network of the Ministry of Health shows that Enterobacteriaceae occupy a more important position in bacterial resistance monitoring,and it is also necessary to pay attention to Enterobacteriaceae in the monitoring of bacterial resistance of livestock and poultry sources.For the grassroots farms,they cannot obtain the data related to the monitoring of bacterial resistance of livestock and poultry sources,and cannot scientifically apply the results of drug resistance monitoring.Therefore,monitoring the drug resistance of Enterobacteriaceae bacteria of livestock and poultry origin and developing a drug resistance monitoring management system for livestock and poultry farms are important for the subsequent research and control of bacterial resistance of livestock and poultry origin.1.The isolation and identification of Enterobacteriaceae bacteria in Huabei,Anhui ProvinceThe Enterobacteriaceae bacteria isolated in this project came from the manure sent from livestock and poultry farms in Huabei,Anhui Province.After isolation and purification by Erythromax Blue agar medium(EMB)and Mac Conkey agar medium(MAC),microscopic examination by Gram staining and biochemical test,and identification by sequencing with16 S r RNA primers,a total of 12 strains of E.coli of porcine origin,9 strains of E.coli of chicken origin,5 strains of Klebsiella pneumoniae of porcine origin,1 strain of Klebsiella pneumoniae of chicken origin,3 strains of Aspergillus chimaericus of porcine origin and 4strains of Aspergillus chimaericus of chicken origin were isolated.and 4 strains of Chlamydomonas reinhardtii of chicken origin.2.Drug resistance of Escherichia coli,Klebsiella pneumoniae and Aspergillus chimaeraAccording to the type of drugs,14 commonly used antibiotics were selected,and the drug susceptibility of 34 strains of Enterobacteriaceae was tested by the production agar diffusion method(K-B method),and the results showed that all 34 isolates were resistant to penicillin and ampicillin,among which the resistance rate of Escherichia coli of porcine origin to 7 antibiotics exceeded 60.00%;the resistance rate of Escherichia coli of chicken origin to 9 antibiotics exceeded 60.00%;The resistance rate of Klebsiella inflammatoryis to 10 antibiotics exceeded 66.67%;the resistance rate of Heterotrophomonas pigens to 13 antibiotics exceeded 66.67%.And all the isolated strains showed different degrees of resistance to 14 antibiotics,which showed that the multi-drug resistance was more serious.The PCR assay technique was used to detect 16 drug resistance genes in 34 isolated Enterobacteriaceae.the PCR assay results showed that for 16 drug resistance genes E.coli could be amplified,among which the highest detection rate was TEM(80.95%)and the lowest was arm A(4.76%)and tet C(4.76%);Klebsiella pneumoniae did not detect CTX-M,arm A,qnr B,tet C genes were not detected in Klebsiella pneumoniae,and the resistance genes with detection rates over 50.00% were OXA,ant(3’’)-Ia,EM,qnr D,qnr S;rmt B,arm A,qnr S,qnr B,tet C genes were not detected in K.oddis,and the detection rates over50.00% of drug-resistant genes were OXA,ant(3’’)-Ia,qnr D,SHV(57.14%),aac(6’)-Ib(57.14%).3 Development of a drug resistance monitoring management system for livestock and poultry farmsIn order to assist the management personnel to observe and interfere with the generation of drug resistance,the livestock and poultry farm drug resistance monitoring management system is used to manage and analyze the drug resistance situation,antibiotic use and elimination situation on the farm,and to understand the number and development trend of drug resistant bacteria on the farm,so as to help develop a system to control drug resistance and guide the use of drugs.Therefore,this system develops functions such as drug resistance detection management,veterinary drug inlet and outlet management,veterinary diagnostic drug record management,veterinary drug usage management,and extermination management for the management of drug resistance situation and factors influencing drug resistance generation in livestock and poultry farms.This topic uses the drug resistance of some Enterobacteriaceae bacteria in livestock and poultry farms as the basic data for the drug resistance detection management of the drug resistance monitoring management system in livestock and poultry farms,and provides support for the management analysis of the subsequent system.In summary,this study took 12 strains of E.coli of pig origin,9 strains of E.coli of chicken origin,5 strains of Klebsiella pneumoniae of pig origin,1 strain of Klebsiella pneumoniae of chicken origin,3 strains of Methylobacterium oddum of pig origin and 4strains of Methylobacterium oddum of chicken origin isolated from livestock and poultry farms in Anhui Province as the research objects,and found that Enterobacteriaceae bacteria in Anhui Province have a certain degree of resistance to commonly used antibiotics,among which E.coli carried the most resistant genes,Klebsiella pneumoniae and Aspergillus oddus carried relatively few resistant genes,but the resistance of Aspergillus oddus was more serious.The drug resistance monitoring management system for livestock and poultry farms was also developed to provide data support and visualization ways to observe and interfere with the generation of drug resistance in livestock and poultry farms. |
