Abstract/Summary:
Optical therapy is a new type of tumor therapy,which uses a specific light source to irradiate optical therapy drugs enriched in the tumor site to produce photothermal effects,active oxygen species,etc.,so as to kill tumors.Compared with traditional tumor treatment,optical therapy has the characteristics of strong targeting and less trauma.R-780 derivatives are a class of small molecule flower dyes with good broad-spectrum tumor targeting,and human serum albumin is an excellent carrier for drugs.We investigated whether the drug could perform efficient aggregation at the tumor by means of human serum albumin as a carrier to solubilize IR-780-F synthesized based on IR-780,and whether the drug could be efficiently aggregated at the tumor through blood circulation by tail vein injection,and injected in the tail vein for about 48 hours It was determined that the drug could achieve a large amount of aggregation at the tumor site,which verified the useful and strong tumor targeting ability of human serum albumin solubilization IR-780 and IR-780-F in human serum albumin.By exploring the ability of IR-780-F to generate reactive oxygen species under 808 nm laser irradiation,the correlation between the power density of laser irradiation and the ability to generate active oxygen species was analyzed.The ability of IR-780-F to produce reactive oxygen species in cells further demonstrates the possibility of IR-780-F in photodynamic therapy.After in vivo targeted studies proved that the system can accurately locate tumors,we used tail vein injection in mice to carry out photothermal therapy experiments,when irradiated with 808 nm laser,the temperature at the tumor can reach more than 60 °C,after the experiment,we found that human serum albumin was solubilized IR-780-F can effectively inhibit tumor growth,and after the analysis of mouse tissues after the end of treatment,it was found that the animal organs of the experimental group were intact,which confirmed the biocompatibility and safety of the drug 。 By exploring the photostability of human serum albumin solubilized IR-780-F and human serum albumin solubilized IR-780,the in vivo photostability of IR-780-F was determined to be stronger than that of IR-780.Subsequently,mouse experiments proved that human serum albumin solubilization IR-780-F can achieve tumor inhibition by multiple photothermal treatments with one injection.This article explores the tumor targeting ability,photothermal therapy ability and photodynamic therapy ability of using human serum albumin solubilizing IR-780 derivatives and reducing DMSO content to 1%,which lays a solid foundation for the next step to enter clinical trials and explore IR-780-F in photothermal/photodynamic combination therapy.
