Electrochemiluminescence(ECL)is a kind of light radiation produced by the chemical reaction between the electrode reaction products or between the electrode reaction products and some components in the solution by applying a certain voltage on the electrode.ECL has the advantages of both electrochemistry and chemiluminescence,such as high sensitivity,low detection limit,wide linear range,a simple instrument,convenient operation,easy automation,and so on.ECL is a new detection technology,which can not only provide luminous intensity and electrochemical signals but also provide more information about optical detection and imaging.Recently,the development of this technology shows its excellent ability in detecting microscopic biological interfaces,for example,many biological processes are related to ionic membranes.In this paper,through the construction of a membrane-supported porin system,the application potential of ECL in ion channel and membrane activity analysis was explored.(1)In this experiment,a suitable ECL membrane system based on tetraoctylammonium bromide was chosen,and the reaction of halogen ions to ECL was investigated.Firstly,the binding strength of halogen ions was analyzed by the electrochemical method,and the results were consistent with the Langmuir isotherm.Then,the dynamics of anions across the membrane are obtained from the time-dependent ECL statistical data,and the ECL signal is accurately located by the mass transfer in the orientation of the nanocages.Finally,the mechanism of ion transport was determined by halogen selective fluorometry.This analysis strategy may replace the previous nanopore and fluorescence analysis methods,and become a new ECL application for various surface-constrained transient scene measurements,such as in-vitro gated ion or molecular transport.(2)Based on zinc porphyrin organic cage(Zn PC),tetraoctylammonium bromide was replaced by phospholipid of Biological cell membrane.Lipophilic Zn PC is a unique geometric hollow structure constructed by dynamic covalent chemistry.Its aliphatic vertices make it have some gaps and easily fuse with biomembrane.ECL is insensitive to H2O2 due to the properties of the lipid bilayer.It is found that the system composed of phospholipids,cholesterol,and the organic cage is a kind of ECL system sensitive to the membrane,which is converted into visible ECL signal by the influence of Ca2+and piercing peptides on the membrane structure on the microscopic level.At the same time,the vesicles were prepared by the system,and the vesicles were incubated with the cells,and the vesicles were imaged.
