Autism Spectrum Disorder(ASD)is a wide range of neurodevelopmental disorders.Patients are often accompanied by symptoms such as enteritis,constipation and diarrhea,and their intestinal symptoms are closely related to behavioral problems and emotional disorders.Bifidobacterium lactis Probio-M8 isolated from healthy women’s breast milk,has been proved to have good gastrointestinal digestive tolerance,anxiety relief and Alzheimer’s disease symptoms.In this study,38 C57BL/6N mice were used as experimental animals,Prevent group:the female mice were given Bifidobacterium lactis Probio-M8(2×10~8CFU/ml)2 weeks before the model was established for early prevention.;Treat group:the newborn mice with ASD were given Probio-M8(2×10~8CFU/ml)for 6 weeks after weaning;Model group and Normal group,normal saline was given.Taking behavioral experiment,neurotransmitters,intestinal flora and its predicted metabolites as indicators,the effect of Bifidobacterium lactis Probio-M8 on ASD model mice.1.Establishment of ASD model mice by valproic acid(VPA):VPA modeling does not affect the growth of mice,and ASD mice show autistic behaviors such as social disorder,anxiety,learning and memory decline,which indicates that the ASD model mice has been successfully established.2.Behavioral experimental results:In the three-box social experiment,open field experiment and new object recognition experiment,the social ability,anxiety,repetitive rigidity and learning and memory ability of the Prevent and Treat mice(6-9 weeks)were improved.3.Analysis of intestinal flora:There are 8 phyla,51 genera and 90 strains in 114 feces samples of mice.Through the analysis ofαandβdiversity,the abundance and structure of intestinal flora of Prevent and Treat mice were significantly improved.Through the analysis of strain difference,The contents of Bifidobacterium globosum,Lactobacillus johnsonii and Akkermansia muciniphila,the beneficial bacteria in the intestines of the mice of Prevent and Treat,increased significantly,while the contents of Bacteroides acidophilus decreased.4.Analysis of neurotransmitters in blood:The contents of 5-hydroxytryptamine,tryptophan and succinic acid in blood of the Prevent and Treat mice increased significantly(P<0.05),and the contents of glutamic acid in the Treat mice increased significantly(P<0.05).Prediction of metabolites in feces:Cholesterol of Prevent and Treat mice decreased,while lithocholic acid and phenylacetate of Prevent mice increased significantly(P<0.05).And methylxanthine of Model and Prevent mice increased(P<0.05).In summary,information exchange,anxiety,repetitive stereotypes and memory ability of ASD model mice were improved by probiotics intervention.The abundance and beneficial bacteria of intestinal flora increased,the content of 5-hydroxytryptamine increased,the content of cholesterol in metabolites decreased,and the contents of lithocholic acid and phenylacetate increased,which maintained the steady state of metabolic system.This research can not only broaden the research field of intestinal microbes and human health,but also provide theoretical basis for the improvement of intestinal symptoms of autistic children.
