Sugar beet(Beta Vulgaris L.)is an important sugar crop in China and an important cash crop in Inner Mongolia.At present,drought,low temperature and soil salinization are important environmental factors affecting the growth and development of sugar beet,which seriously restrict the high quality development of sugar beet production.ERF(Ethylene-resp onsive element binding factors)are important transcription factors regulating plant growth and development and responding to stress,and play an important role in plant response to biotic and abiotic stresses,regulating.They play an important role in plant response to biotic and abiotic stresses,in regulating the expression of stress-related functional genes,and in improving plant stress resistance.Transcriptomics-based studies have shown that some sugar beet ERF transcription factors play important roles in sugar beet tuber development and sugar accumulation.In this study,we screened and cloned the beet ERF transcription factor Bv_ammr gene,systematically investigated its expression pattern under abiotic stress and hormone induction,and genetically transformed it to analyze its preliminary function,so as to provide a theoretical basis for further elucidating the molecular response mechanism of ERF transcription factor genes in the regulation of sugar beet tuber development and stress of adversity.The main findings are as follows:(1)The complete CDS region of Bv_ammr gene was cloned from sugar beet.Bioinformatics analysis showed that the CDS region of Bv_ammr gene was 906 bp in length and encoded 301 amino acids.The molecular weight of the protein was 33.19 k Da,the theoretical isoelectric point was 8.61,and the secondary structure of the protein was mainly random curly and α helix.It had a conserved AP2 domain,but no transmembrane domain.The homology of amino acid sequence and evolutionary tree analysis showed that the protein was closely related to aristolochia pili.(2)The protein encoded by ERF transcription factor is mainly localized in the nucleus,and the yeast single hybridization experiment demonstrated that it has a reciprocal relationship with the GCC-box,which has transcriptional activation activity.(3)The Bv_ammr gene is mainly expressed in the late stage of sugar beet reproduction,and it is mainly expressed in the root system in the high-yielding varieties,while it is mainly expressed in the leaves in the high-sugar varieties.(4)The Bv_ammr gene responded differently to abiotic stress and hormone induction,significantly for thephon and salicylic,and genetic transformation of Arabidopsis showed that the gene had growth-promoting effect,. |
