We extracted the total DNA samples of lichen biological soil crusts from the surface of laoheishan volcano in Wudalianchi,analyzed their physical and chemical properties,used the method of metagenomic sequencing to analyze the microbial community composition,and statistically analyzed the experimental results with image tools and statistical software to explore the relationship between the composition of lichen crust microbial community and environmental factors in the ecological distribution of Laoheishan volcano.And focus on excavating the microbial species resources that participate in the cycle of iron and sulfur to provide important potential,and analyze and predict the metabolic pathways and functional genes of iron and sulfur in Laoheishan volcanic rock and geomorphology microorganisms.The results of this paper are as follows:(1)The iron conversion rate of the lichen crust samples was:0.00588μmol/(g·d),and the sulfur conversion rate of the lichen crust samples was:0.05986μmol/(g·d).(2)In this study,the microbial diversity and OTU taxonomic distribution of lichen crust in laoheishan volcanic rocks showed that Actinobacteria,Verrucomicrobia,Gemmatimonadetes,Planctomycetes and armored bacteria Armatimonadetes were more abundant.The data showed that the microbial diversity index of iron-treated lichen crusts was about 2.88,and that of sulfur-treated lichen crusts was 2.94.The microbial diversity of sulfur-treated lichen crusts was better than that of iron-treated lichen crusts.The lichen crust samples before and after iron-sulfur treatment were sequenced and analyzed,and it was found that the Wudalianchi Laoheishan lichen crusts had rich flora species.In the iron treatment group,Pantoea,Actinoplanes,Burkholderia and Alternaria were the main dominant bacteria;in the sulfur treatment group,Sphingomona,Lichenibacterium,Enterobacter,Pantoea,Alternaria,Acidisphaera,Kosakonia and Cronobacter are the dominant bacteria.(3)Iron cycle-related genes(feo A,cyc A,dps),ferrous oxidase(iro),ferritin(ton B,exb B,exb D)and cytotoxins(mbt E,fyu A)involved in the iron cycle before and after iron treatment in Wudalianchi old Heishan crustaceans,cells Pigment(cox A)absorbs iron;(pet A,pet B)realizes photosynthesis in the process of iron absorption.Fe2+(feo A,feo B),iron-containing outer membrane transporter(flu),and ABC transport system(exb D,dps,cys A)express iron transport;dps and bfr store iron in the form of stationary phase nucleoid ferritin.Sulfur cycle-related genes cys J,cys NC,cys I,cys D,cys H,cys N,cys C and sir involved in the crustaceans of Wudalianchi before and after sulfur treatment were assimilated sulfate reduction genes;sor A and sor B were sulfur oxidation genes;cys P,cys A,cys W,cys U are sulfate transport genes;apr A,apr B are dissimilatory sulfate/reduction genes.The sulfur cycle related genes cys J,cys NC,cys I,cys D,cys H,cys N,cys C,sir,sor A and sor B,which were involved in the sulfur treatment of the Lichen crust in Laoheishan,Wudalianchi,were assimilated sulfate reduction genes;cys P,cys A,cys W and cys U are sulfate transporter genes,and apr A and apr B are dissimilatory sulfate/reduction genes.cys J,cys H,cys C were Letharia,cys NC were Acidisera,cys I cys D,cys N and sir were Cabonia,Mucilaginibacter,Acidisphaera,Frankia,sor A,sor B were Mesorhizobium,Acetobacter;The microbes of cys A,cys W,cys U,apr A and apr B were Massilia,Pantoea,Massilia,Hyphicrobiaceae,Alphaproteobacteria.
