The formation of the mammalian cortex is essential for the development of nervous system.Cerebral cortex is crucial for the fundamental function of physiology and metabolism process.Meanwhile,fot the high level functions of the brain,such as the cognition,learning and memory,the regulation of the cortex is also very importantant.The stability of m RNA is particularly important when it is transferred to the cytoplasm from nucleus.HuR,one of the RNA binding protein,maintains the stability of thousands of m RNAs in cytosol.In order to explore the role of HuR in neural development,we constructed the HuR conditional knockout mice by Cre-Lox P system.Which could inhibit the expression of HuR by specifically knockout the HuR gene in the cerebral cortex neurons.After confirming the spatial and temporal expression of HuR,wepreformed the behavioral tests by wt and HuR conditional kockout mice to see if the deletion of HuR will cause abnormal behaviors for mice.For the mice behaviors test,we carried out open field,light-dark box,elevated plus maze and morris water maze.In mammals,the normal function of nervous system functions is required for maintaining life activities.At different neural development stages,abnormal development of the nervous system can lead to dysfunction brain and abnormal behaviors.Therefore,we expect to clarify the relationship between the abnormality of gene expression and behavior change through our study.Through the test analysis,we found that the knockout mice in the light-dark box,elevated plus maze,olfactory test,novel object recognition test and morris water maze test have no obvious anomalies,but in open field experiment,the HuR knockout mice displayed preference to staying longer in the central area.And at the same time in the social interaction experiments we found that HuR knockout mice spent more time in following and push-crawl with each other.In this study,we found that HuR is very important for neural development,especially in the formation of cerebral cortex.The main result is that the lack of HuR promotes the social behavior of the mice,which lays a foundation for the future research on the specific molecular mechanism of HuR in regulating neural development.As an RNA binding protein,HuR may regulate the assembly process of the cerebral cortex by maintaining the stability of target m RNA in the nervous system,thus affecting the social behavior of animals.
