In the fields of biomedicine,energy chemical industry,material metallurgy,etc.,the particle size,morphological characteristics and other indicators have an important influence on the quality and performance of the product,and particle analysis has become an indispensable key link in the research process.In order to realize the full inspection of particle morphology in biomedicine,this paper designs a method of image acquisition and analysis of flowing particles based on microscopic imaging.In view of the fact that the number of particles in the images collected each time is different and the falling position is random when the particles are in a flowing state,the dynamic image definition evaluation parameters and focusing control algorithm are studied,and the acquisition of the particle image in the flow cell with a depth of 80?m is completed.Different evaluation functions are used to analyze the static particle image,and the Brenner function suitable for the particle image is determined as the sharpness evaluation function.Then it is applied to the flowing particle image,combined with the imaging characteristics of the flowing particle image,the image clarity evaluation curve obtained before and after is compared,the deficiencies are analyzed and the flowing particle image is processed accordingly.In the experiment,a suitable threshold is selected to segment the flowing particle image to obtain a binary particle image,and the opening and closing operation of mathematical morphology is used to enhance the target particle in the image.A single particle image is obtained according to the particle extraction process,and on this basis,the definition of the entire image is evaluated,and finally a feasible method for evaluating the definition of flowing particle image is obtained.By analyzing the automatic focusing process of the flowing particle image,a local interval traversal search algorithm is proposed to avoid the interference of local extrema on focusing.The initial position and focusing interval of the system during focusing are determined through experiments,and the driving motor drives the microscope objective to move quickly to the vicinity of the coarse focal plane.Control the working state of the motor according to the automatic focusing algorithm,search with the best step length obtained from the experiment,combine the image clarity evaluation value to determine and return to the best imaging focal plane and then stop moving,and the computer saves the collected clear particle image.Through the selection of optical imaging components,a flow particle image acquisition device was built.The matching process of the camera acquisition frequency,gain,exposure time and light source brightness was studied,and the image acquisition parameters were set for automatic focusing,and clear images of flowing particles were obtained.Finally,a system calibration and verification experiment based on standard particles is designed,standard particles of different sizes are used for automatic focusing experiments under the same experimental conditions,the experimental results obtained by the method given in this article are compared with actual data,the average error of target particle size is less than 1?m,which shows that the flow particle image acquisition system and automatic focusing method constructed in this paper are effective and reliable.
