Purpose:Explore the effect of Wenqingyin on the inflammatory factors of acute anterior uveitis induced by endotoxin in rats.Material and method:In this experiment,30 SD rats will be selected,half male and half male,and the rats will be randomly divided into 6 groups,5 rats in each group,which are the blank control group(K),model control group(M),and positive drug control group.(Y)and Wenqingyin administration group(low concentration group: 0.25g/ml,medium concentration group: 0.5g/ml,high concentration group: 1g/ml).The Wenqingyin administration group was given Wenqingyin(0.25g/ml,0.5g/ml,1g/ml)by gavage for 21 days.The model control group was given the same volume of distilled water by gavage for21 days.Except for the blank control group,the other five groups were injected with endotoxin(20?l/mouse)under the sole of the feet after intragastric administration for 7 days and 14 days,respectively,to establish an animal model of acute anterior uveitis in SD rats.Slit-lamp observations were performed on the anesthetized rats before administration and after modeling,and inflammation scores were given.After 21 days of intragastric administration,the rat eye tissues were taken out for pathological observation,paraffin pathological sections and frozen pathological sections were made,and hematoxylin and eosin(HE)staining method was used to detect the histological changes of the iris and ciliary body of each group.The anterior chamber Internal fibrin membrane accumulation and inflammatory cell infiltration;immunofluorescence staining was used to detect the expression of inflammatory factors(IL-1?,IL-6,TNF-?)in the iris and ciliary body of rats in each group.Results:1.Rat anterior segment inflammation performance and score: Compared with the blank control group(K),the model control group(M)showed significant inflammation in the anterior segment(P<0.001);the positive drug control group(Y)was similar to the model control group Compared with,the inflammatory reaction was significantly reduced(P<0.001);the Wenqingyin low-concentration group(D)was not statistically significant compared with the model control group(P>0.05)the Wenqingyin medium-concentration group(Z)(P<0.05)and Wenqing Compared with the model control group,the high-concentration drink group(G)(P<0.01)had reduced inflammation.2.Histopathological observation of the iris and ciliary body: Compared with the blank control group,the model control group has significantly dilated iris blood vessels,thickened iris edema,and a large amount of fibrin membrane accumulation and inflammatory cell infiltration in the anterior chamber;positive drugs Compared with the model control group,the control group has significantly reduced iris vasodilatation,the iris morphology tends to be normal,and the fibrin membrane accumulation in the anterior chamber and the infiltration of inflammatory cells are significantly reduced;the low-concentration group of Wenqingyin is not significant compared with the model control group Compared with the model control group,the iris vasodilatation was reduced,fibrin membrane accumulation and inflammatory cell infiltration were reduced in the Wenqingyin medium concentration group and the Wenqingyin high concentration group respectively.The iris morphology of the Wenqingyin high concentration group tended to normal.3.The expression of IL-1?,IL-6 and TNF-? in the iris and ciliary body respectively:Compared with the blank control group,the expression of inflammatory factors in the model control group increased significantly(P<0.001);the positive drug control group Compared with the model control group,the expression was significantly reduced(P<0.001);the Wenqingyin low-concentration group was not statistically significant compared with the model control group(P>0.05);the Wenqingyin medium-concentration group(P<0.05)and Wenqingyin high Compared with the model control group,the expressions of the concentration group(P<0.01)were reduced.Conclusion:1.The medium and high concentrations of Wenqing Decoction can effectively reduce the inflammatory manifestations of acute anterior uveitis induced by endotoxin in SD rats.2.The medium and high concentrations of Wenqingyin can effectively reduce the expression of inflammatory factors IL-1?,IL-6 and TNF-? in the iris and ciliary of acute anterior uveitis induced by endotoxin in SD rats.3.Compared with the high concentration of Wenqingyin,the high concentration of Wenqingyin in the EIU model can more effectively reduce inflammation and reduce the expression of inflammatory factors IL-1?,IL-6 and TNF-?.
