Purpose:The method of high fat and high sugar feed combined with injection of STZ was used to establish type 2 diabetic renal damage(diabetic nephropathy,DN)animal model,were interventional treated with moxibustion combined with Chinese medicine sticking Shenque acupoint and internal administration formula(Jin Kui Shenqi Pills and Wuzi Yanzong Pills),for interventional treatment of DN model rats,and treated as a single internal formula(Jin Kui Shenqi Pills and Wuzi Yanzong Pills)and single topical god threshold point of comparison,through testing model rats blood glucose,creatinine(Scr),urea nitrogen(BUN),blood lipid and protein immunoblot(Westem Blot,WB)method to detect transformation in renal tissue growth factor(TGF-?1)the content of protein and fiber connection(FN),drug moxibustion combined with treatment of DN rat renal damage effect and its possible mechanism,to provide emotional support for clinical TCM as a treatment for DN.Material and method:Fifty clean-grade male rats with body weight of 250g±20g were fed adaptively for 7 days.They were randomly divided into 10 rats in the blank control group which were fed with conventional chow and 40 rats in the modeling group which were fed with high fat and high sugar chow.All of them were kept at suitable room temperature for 28 days.Rats with body weight less than 340±10g were excluded from the model group.Before modeling,the rats in the model group were intraperitoneally injected with 50mg/kg STZ after fasting for 12 h.After 72 h,fasting blood glucose(FBG)was measured at 16.7MMOl/L,which was considered as successful modeling and included in the observation.Those whose blood glucose did not reach the standard were given STZ once.The blank control group was injected with the same dose of sterile citrate buffer and continued fed with normal feed,while the modeling group continued to be given high-fat and high-sugar feed and fed with free access to water.Eight rats were randomly selected from blank control group and model group,and blood was collected from orbital vein.The quantitative determination of Scr.BUN by automatic biochemical analyzer was statistically significant(P<0.05),indicating the occurrence of renal damage in rats.A total of 12 animals died due to body weight or low blood glucose or intolerance during the modeling period.Rats in the model group were randomly divided into model control group,internal treatment group,external treatment group and moxibustion combined group,with 7 rats in each group.Internal treatment group was treated with 4Ml /kg/d gavage of Jinkui Shenqi pills and Wuzi Yanzong pills combined formula plus or minus decoction;In the external treatment group,aconite,cinnamon and other drugs were used to grind rice wine and mix and paste Shenthreshold acupoint,once a day;The combination group of moxibustion medicine is the combination of two treatment methods;Blank control group and model control group were intragastrically given the same volume of normal saline at the same time every day.Each group was treated for a course of 7 days,a total of 2 courses.After treatment,blood glucose,blood lipid and renal function(Scr,BUN) of the rats were measured,and the expression of conversion factor ?1 and FN protein in renal tissue was detected by Western Blot.Results:1.Effects on general symptoms of rats: the rats in the model group were weak and weak,emaciated,slow in activity,dull color,and gradually increased diet and water intake.After medication treatment,the above symptoms were significantly improved,but compared with the internal treatment group and external treatment group,the mental state,hair color and body flexibility of rats in the combined moxibustion group were better.2.Effect on FBG in rats: After treatment,compared with the blank control group,the blood glucose in the internal treatment group,external treatment group and internal and external combination group was significantly higher than that in the blank control group Compared with blank control group and significantly decreased(P<0.01);Compared with model control group,internal treatment group,external treatment group,The blood glucose content in the internal and external combination group was significantly decreased(P<0.05);There was no significant difference between internal treatment group and external treatment group(P>0.05).3.Effect on blood lipid of rats: compared with blank control group,TC,TG and IDL-C of model control group were significantly increased(P<0.05);Compared with model control group,TC,TG,LDL-C in treatment groups were significantly decreased,especially in moxibustion combined group(P<0.01).Compared with the blank control group,the content of HDL-C in the other four groups was significantly decreased(P<0.05).4.Effect on Scr,BUN of rats: Compared with blank control group,renal function of model control group was significantly increased(P<0.05);Both Scr and BUN in the treatment group were lower than those in the model control group,and the difference was statistically significant,especially in the moxibustion combined group(P<0.01).There was no significant difference between internal treatment group and external treatment group(P>0.05).5.TGF-?1 and FN in renal tissue were detected by Western-blot method: compared with blank control group,TGF-?1 and FN expression levels in model control group were increased,and gray value was statistically significant(P<0.01);Compared with the model control group,the expression levels of TGF-?1 and FN in the internal treatment group,external treatment group and internal and external combination group were significantly decreased(P<0.01),with statistical differences.There was no significant difference in TGF-?1 and FN expressi on contents between the internal treatment group and the external treatment group(P>0.05).However,compared with the internal treatment group and the external treatment group,the expression contents of TGF-?1 and FN factors in the moxibustion combination group were significantly decreased(P<0.05).Conclusion:1.Moxibustion combined with intervention therapy can change the general state of DN rats and improve part of the pathological symptoms of early renal damage in diabetic nephropathy.2.Moxibustion combined with moxibustion can reduce the content of blood sugar,blood fat and Scr,BUN,regulate the disorder of sugar and lipid metabolism,dispel phlegm and promote blood circulation,remove blood stasis and remove collaterals.3.Moxibustion combined with moxibustion can down-regulate the protein expressions of TGF-?1 and FN,inhibit the progression of glomerular and renal fibrosis,reduce the high glomerular filtration rate,improve renal function,and effectively slow down the development of DN.
