Purpose: Huoxue Qushi Granules are commonly used prescriptions for the clinical treatment of nonalcoholic steatohepatiti(NASH)by Professor Lu Bingjiu.In this study,the NASH mouse model was used as the experimental object,and Huoxue Qushi Granules were used as intervention methods to explore the therapeutic effect and mechanism of Huoxue Qushi Granules on the NASH mouse model.Material and method: The high-fat diet induction method was used to establish the NASH mouse model,and the pathological state of the mouse liver tissue was used as the evaluation standard to verify the model.In the traditional Chinese medicine group,mice in each group were given low,medium and high different doses of Huoxue Qushi granules,and mice in the Western medicine control group were given Yishanfu.The administration was continuously administered for 4 weeks.By comparing with the blank group and the model group,observe the effects of different doses of Huoxue Qushi Granules on the general condition,liver index,liver function and blood lipid indexes,and liver tissue pathology of NASH mice to determine the effect of Huoxue Qushi Granules.The therapeutic effect of wet granules.Based on the STING-IRF3/NF-?B signaling pathway,Western Blot and RT-PCR were used to detect NF-?B p65,p-NF-?B p65,STING,TBK1,IRF3 protein and IL-before and after the administration of Huoxue Qushi granules.6.The expression differences of IL-1?,TNF-?,NF-?B p65,STING,TBK1,IRF3 genes,to explore the mechanism of action of Huoxue Qushi Granules in the treatment of NASH.Results:1.Successfully constructed NASH mouse model.2.Huoxue Qushi granules can improve the general condition,liver index and pathological state of mice.Among them,the improvement was more obvious in the high-dose group of Huoxue Qushi Granules.3.Compared with the blank group,the serum ALT and AST levels of mice in the model group were significantly increased,and the difference was statistically significant(P<0.05);compared with the model group,the high,medium and low dose groups of Huoxue Qushi Granules and western medicine The serum ALT and AST levels of mice in the control group decreased,and the differences were statistically significant(P<0.05);compared with the western medicine control group,the serum ALT and AST levels of mice in the high and medium dose groups of Huoxue Qushi Granules Significantly decreased(P<0.05).Compared with the model group,the serum TG,TC,and LDL levels of mice in the high,medium,and low dose groups of Huoxue Qushi Granules and the western medicine control group were significantly reduced(P<0.05),and the serum HDL levels of mice were significantly increased(P<0.05),among which the high-dose group improved the most.4.Compared with the blank group,the expression of IL-6,IL-1?,TNF-?,and NF-?B p65 genes in the model group increased significantly(P<0.05),but after treatment,their expression decreased Significantly(P<0.05);Compared with the blank group,the expression of p-NF-?B p65 protein in the liver tissue of the model group was significantly increased(P<0.05),and the expression of NF-?B p65 protein was significantly increased(P< 0.05),and after treatment,its expression decreased;compared with the blank group,the expression of STING,TBK1,and IRF3 genes in the model group increased significantly(P<0.05),and the expression of STING protein and its signal pathway downstream The protein expression of IRF3 and TBK1 was significantly increased(P<0.05),and after treatment,their expression decreased significantly(P<0.05).Conclusion:1.Huoxue Qushi Granules can treat NASH by reducing the liver index of mice,improving the liver function of mice,regulating blood lipids and improving liver pathology.2.Huoxue Qushi Granules can inhibit inflammation by down-regulating the expression levels of IL-6,IL-1?,and TNF-? inflammatory factors in the liver.3.Huoxue Qushi granules can enhance the anti-inflammatory reaction ability of liver tissue in mice and inhibit liver inflammatory reaction.The possible mechanism is to regulate the NF-?B signaling pathway.4.Huoxue Qushi Granules can reduce the inflammatory response in liver tissues of mice,regulate blood lipid levels and improve liver injury to achieve the purpose of treatment.The possible mechanism is that Huoxue Qushi Granules regulate the STING-IRF3 signal transduction pathway.5.According to the comparison results of various indicators,the high dose of Huoxue Qushi Granules has better effect than the middle and low doses.
