Purpose:To observe the effect of Huoxue-Qushi Formula for the changing in the content of serum of ALT,AST,TG and LDL, expression of the liver homogenate ICAM-1and IL-1β in the NAFLD model rats, and explore the efficacyand mechanism of drug action. Material and method:In this experiment, the NAFLD models were established by adopting the high-fat diet to feed rats,and simulate the NAFLD process.Sixty rats were randomly divided into model group(50 rats) and blank group(A group,10 rats),model group were fed with high-fat diet,blank group were fed with general diet,starting from the forth week,one rats in model group were terminated and treated by liver HE staining at every weekend randomly,the NAFLD models were established at the eighth weekend. The model group were randomly divided into model group(A group),the high,medial and low-dose group of Huoxue-Qushi Formula(C,D and E group) and silybin group(F group) with nine in each.Starting from ninth week,the model group were fed with general diet instead,others were with drug intervention.The twelfth weekend,while after twelve hours of fasting,the rats were terminated after weighting and taking material,weighting the liver,determining the content of serum of ALT,AST,TG and LDL,observing the change of the liver homogenate ICAM-1 and IL-1β by using RT-PCR and western-blot. Results: 1.General condition:the model group were much more fatter, slower,more depressed,loss of appetite than blank group,after four weeks with drug intervention,blood group symptoms improved significantly. 2. HE staining:the model group showed moderate to severe hepatic steatosis, lobular inflammatory cell infiltration, there are more focal fibrosis; each treatment group liver tissue steatosis degree improved significantly compared with the model group, wherein the high-dose group and the control group basically close, low, medium dose group showed signs of improvement, Capsules and group recovered. 3.Compared with the blank group, The ALT, AST, TG and LDL of the model group increased obviously(p <0.05 or p <0.01); Compared with the model group, The ALT, AST, TG and LDL of the all dose Huoxue-Qushi Formula group decreased distinctly(p<0.05). And the high dose group decreased significantly than others. 4.Compared with the blank group, the ICAM-1 and IL-1β expression of the model group increased obviously(p <0.05 or p <0.01);Compared with the model group,the ICAM-1 and IL-1β expression of Huoxue-Qushi Formula high dose group reduced significantly(p<0.01); also found that the ICAM-1 and IL-1β enhanced expression were positively correlated with their pathological changes. Conclusion: 1.The liver injury caused by high-fat diet, Huoxue Qushi Formula can significantly reduce the weight and hepatic index of rats,reduce liver lipid deposition,have antiobesity effects in some degree. 2. Huoxue Qushi Formula can significantly reduce the ALT、AST、TG and LDL, protect liver cells,reduce inflammation of liver cells and play the an important role of prevention and treatment of NAFLD;The high-dose group treatment was better than low and middle dose group. 3. The high ICAM-1 and IL-1β expression of liver tissue may promote and take part in the fat-induced liver injury; For Huoxue-Qushi Formula,the one of the action mechanisms may restrain the ICAM-1 and IL-1β expression to reduce inflammation and liver cell damage, so as to improve function,reduce blood fat,treatment of non-alcoholic fatty liver.
