The prevalence of osteoarthritis(OA)is constantly increasing with age.Mesenchymal stem cells(MSCs)have emerged as a promising approach to treat OA.Adipose-derived(AD-)and umbilical cord-derived(UC-)MSCs are attractive sources in regenerative medcine;however,they have rarely been studied in terms of comparing their effectiveness in chondrogenic ability and OA intervention.In vitro proliferation was analyzed using a cell counting kit(CCK-8)according to the manufacturer's recommendations.Briefly,UC-MSCs and UC-MSCs were seeded into 96-well plates at a density of 2000 cells/well.Ten ?l CCK-8 reagent was added per well at 1,2,3 and 4 days,respectively,and incubated for 2 hours at 37 ?.Optical density(OD)was measured at 450 nm by a microplate reader.The results showed that the proliferation ability of UC-MSCs was significantly higher than that ofChondrogenic differentiation of UC-MSCs and AD-MSCs was induced using a pellet-culture method.Culture-expanded MSCs were trypsinized,neutralized in DMEM/F-12 with 10% FBS.Aliquots of 5×105 cells,resuspended in 1 ml complete chondrogenic medium,were centrifuged at 300 g for 5 min in 15-ml polypropylene conical tubes.Pelleted cells were incubated at 37? under 5% CO2 with loosened caps to permit gas exchange.Within 24 h of incubation,the sedimented cells formed a spherical aggregate at the bottom of each tube.The medium was changed every 3days and at 5 weeks the pellets were harvested.In order to compare the chondrogenic differentiation potential of the two types of cells,frozen sections of Pellet cell clusters were performed,and followed by pathological staining and glycosaminoglycan(GAG)content determination.The results showed that the chondrogenic differentiation potential of UC-MSCs and AD-MSCs was similar.Finally,to compare the retarded effect of UC-MSCs and Ad-MSCs on OA,the anterior cruciate ligament transection(ACLT)surgery was used to induce a rat OA model.The collected joint samples were fixed,decalcified,dehydrated,paraffin embedded and sectioned.Pathology staining,immunohistochemical staining and TUNEL staining results demonstrated that ACLT OA model construction was successful,and intra-articular injection of two kinds of cells significantly improved OA progress,restored the proteoglycan content,inhibited the cartilage cell apoptosis.There was no significant difference between the effects of the two kinds of cells.Interestingly,only a single cell could achieve a good therapeutic effect.
