?Objective? To prove the effect of PPAR? agonist on chondrogenic differentiation of human synovial fluid-derived mesenchymal stem cells(hSF-MSCs)and inhibition of the inflammatory cytokines releasing in inflammatory conditions.?Methods? Synovial fluid was obtained from the ill-knee joints of the patients suffering from knee osteoarthritis by 5m L injection syringe.And then isolating,culturing and identifying hSF-MSCs.Cultured by the Pellet method simulating 3D condition for chondrogenic differentiation,hSF-MSCs were divided into 4 groups:blank control group,TGF-?1group,TGF-?1+GW0742 group and GW0742 group.The effects on hSF-MSCs' proliferation and chondrogenic differentiation of GW0742 were evaluated by comparing the cell proliferation,the cartilage-special marker SOX9 gene expression and protein synthesis and Alcian Blue/toluidine blue staining.Besides,setting 3 different groups(blank control group,hSF-MSCs group,hSF-MSCs+GW0742 group)and detecting TNF-? ?IL-1?expression in each group to estimate whether GW0742 + hSF-MSCs had effects in the aspect of anti-inflammatory ability.?Results?hSF-MSCs were successfully isolated from the knee joints of the patients with osteoarthritis by direct adherence and identified.As for the multiplication capacity,TGF-?1 group,TGF-?1+GW0742 group and GW0742 group had no obvious effects on hSF-MSCs.According to the detection result of SOX9 in PCR and Western Blot,GW0742 group had the most conspicuous effect on chondrogenic differentiation.However,cells in GW0742 group could secrete more aggrecan than TGF-?1+GW0742 group in terms of the section-staining.Furthermore,as for the expression of inflammatory factors,hSF-MSCs+GW0742 group was obviously fewer than hSF-MSCs group.? Conclusion ? PPAR? agonist GW0742 can not only improve hSF-MSCs chondrogenic differentiation,also can enhance anti-inflammatory effect in the inflammatory conditions with hSF-MSCs.It may provide a theoretical basis that PPAR? agonist could have a better treatment with mesenchymal stem cells to osteoarthritis in the future.
