Dendrobium is one of the precious medicinal plants in my country.It has extensive physiological activities,including anti-oxidation,anti-inflammatory,anti-liver injury,anti-tumor and so on.Dendrobium can not only be combined with other drugs in clinical practice,but also can be used as a health care product to strengthen the body and enhance immunity.CYP 450 is a major metabolic enzyme in the liver and is involved in the biotransformation of many substances in the body.After entering the body,the active components of Dendrobium are affected by the metabolism of cytochrome P450 enzyme on the one hand,and the active components themselves also react on cytochrome P450 enzyme on the other hand.Therefore,studying the effect of Dendrobium on cytochrome P450 enzymes is of great significance for ensuring the safety of Dendrobium's clinical combination medication and the safety of taking it as a health care product.Dendrobine and dendrobol are one of the main active ingredients in Dendrobium,which have certain activities in anti-inflammatory,antioxidant,cardiovascular protection and other aspects.This project studies the effects of dendrobine and dendrobol on the main cytochrome P450 enzymes,and provides more theoretical basis for improving the efficacy of dendrobium in clinical use and the safety of its use as health care products.This subject also detects whether dendrobol and dendrobine cause damage to the liver of mice under low,medium and high dosages,which is of great significance for evaluating the safety of dendrobol and dendrobine.The main research content and results include the following parts:1.The effects of dendrobine and dendrobol on the expression of main cytochrome P450 enzymes.In this chapter,C57BL/6 mice were injected intraperitoneally with dendrobine and dendrobiol to study the effects of dendrobine and dendrobiol on the expression of cytochrome P450 enzyme mRNA and protein..The results of mRNA level experiments showed that dendrobine can up-regulate the expression of Cyp1a1,Cyp1a2,Cyp2a5,Cyp2d9 and Cyp3a11 mRNA;Dendrobium can increase the expression of Cyp1a2 and Cyp2b9 mRNA.Protein level experiments show that dendrobine can significantly up-regulate the protein expression of CYP1A1,CYP2 B and CYP2C19;dendrobium can significantly increase the protein expression of CYP1A1,CYP1A2,CYP3 A,CYP2B,CYP2C19 and CYP2E1.2.The effect of dendrobine and dendrobol on main cytochrome P450 enzyme activity in mice.Based on the research basis of the previous chapter,this chapter experimentally studies the effect of dendrobine and dendrobol on the activity of cytochrome P450 enzymes.C57BL/6 mice were administered intraperitoneal injection of dendrobine and dendrobol,and the enzyme activity was used as an indicator to study the effects of dendrobine and dendrobol on CYP1A1,CYP1A2,CYP2 B,and CYP2 C enzyme activities using the substrate probe method.The results show that dendrobine can significantly increase the activity of CYP1A1 enzyme,but has no conspicuous impact on the enzyme activities of CYP1A2,CYP2 B,and CYP2 C.Dendrobol has no conspicuous impact on the enzyme activities of CYP1A1,CYP1A2,CYP2B,and CYP2C.Combined with the experimental results in the previous chapter,it can be seen that when using dendrobium and dendrobium as health care products,special attention should be paid to their use together with other CYP1A1 enzymes that can induce or inhibit the CYP1A1 enzyme.3.Effects of dendrobine and dendrobinol on liver function in mice.Based on the study of the effects of dendrobine and dendrobinol on P450 enzyme,we further studied whether dendrobine and dendrobinol have certain effects on liver function of mice.C57BL/6 mice were selected and intraperitoneally injected with dendrobine and dendrobol to detect the serum and liver related indicators of the mice,and observe the pathological tissue slices of the liver tissue.The experimental results showed that the AST and ALT indexes in the serum and the GSH and MDA indexes in the liver did not show obvious abnormal changes and the pathological sections of liver tissue showed no obvious abnormalities in liver cells.It can be concluded that at the dose used in this experiment,dendrobine and dendrobol have no hepatotoxicity to the liver of mice.
