Dendrobium is one of rare material in China. It has many functions, such as anti- tumor, anti- senescence, reinforcement immunity. So the dendrobium has the wide use in clinical. There are seventy-six kinds dendrobium in China, and the Dendrobium nobile is the most famous one. This assay selected the material of D. nobile which grow in GuiZhou to study. First, we compared the extraction efficacy of total alkaloid in Dendrobium nobile under different extraction conditions, then we confirmed the best conditions to extract. After that, total alkaloid content and main composition (dendrobine) contents were determinated by UV-VisSpectrophotometer and high performance liquid chromatography. Simultaneity, we detected the heavy metal in Noble dendrobium and its grow environment. The results were as follows:1. We compared three kind of extract method found that the transonic is the most effective method. And we optimization this method, conclusive we confirmed the best conditions for this method.2. We determined the total alkaloid contents in Noble dendrobium by detect the absorbance of dendrobine at at the wavelength of 620nm. The calibration curve for dendrobine was linear within the concentration range of 2-20μg/ml, the equation is A=-0.0016+0.0327C(r=0.99984,n=5). And the stability, recovery indicated that this method is dependable for determination the alkaloid contents in Noble dendrobium. The determination results of total alkaloid contents in twelve Nobile dendrobium from Guizhou province is between 0.237-0.480%.3. Select the content of dendrobine in Noble dendrobium as the quality control factor. To establish a method for quantitative analysis of dendrobine in Dendrobium nobile Lindl by high performance liquid chromatographic (HPLC). The equation is A=115936+6387.6C (ng/μl)The determination results of dendrobine contents in twelve Nobile dendrobium from Guizhou province is between 0.073-0.143%.4. The result of heavy metal contented in Noble dendrobium and its grow environment is concern with the amount of total alkaloid. The more heavy metal is the less alkaloid amount is.
