The highland barley and ghee is a special diet in Tibet,in which ?-Glucan in highland barley,unsaturated fatty acids and conjugated linoleic acid in ghee are the bioactive substances that help to maintain intestinal homeostasis.Diet plays a major role in shaping composition of gut microbiota and is closely associated with the occurrence of a variety of diseases,especially intestinal diseases.Inflammatory bowel diseas'e(IBD)is chronic,recurrent and difficult to cure with unknown etiology,among which intestinal flora disorder is the main clinical symptom.We explored the effect of highland barley and ghee diet on gut microbiota in mice,and proposed whether the combined effect of highland barley and ghee diet could improve the intestinal inflammation induced by dextran sodium sulfate in mice.The results showed that the customized highland barley and ghee dietary could adapt the normal nutritional needs of mice,and there was no difference in body weight compared with control group(fed with normal diet).On this basis,the enteritis disease model group was introduced through the three cycles DSS modeling experiment.Compared to control group,the abundance of bacteria was increased and the diversity of intestinal flora,such as Allobaculum,was improved in wild type mice fed on the highland barley and ghee dietary.However,when the mice were treated with DSS,the highland barley and ghee dietary group had high DAI score(p<0.05),showed obvious intestinal ulcers and disorganized intestinal glands by histopathological examination,and decreased intestinal flora abundance and diversity(p<0.05).But the numbers of Clostridium and Sartreella were increased in highland barley and ghee dietary group(p<0.05).In conclusion,the highland barley and ghee dietary improved the intestinal microbial composition and increased intestinal probiotics in mice,but had no effect on DSS induced enteritis.
