Research purpose:Based on the software of traditional Chinese Medicine Inheritance and calculation platform(V3.0),this paper explores Professor Wang Sumei's prescription and medication rules in the treatment of childhood autism spectrum disorder(ASD),and explores the potential mechanism of common drugs in the treatment of ASD by using network pharmacology,so as to guide clinical practice.Research method:1.Collect the medical records and prescriptions of Professor Wang Sumei with the treating ASD of children from January 1,2010 to December 12,2020 in the Dongfang Hospital Beijing University of traditional Chinese medicine.The information was input into the traditional Chinese Medicine Inheritance computing platform.The data mining methods such as association rule analysis and K-means clustering analysis were used to obtain the drug frequency,four Qi,five flavor,meridian tropism,efficacy and prescription rules,to summarize Professor Wang Sumei's treatment ideas about ASD.2.Select Professor Wang Sumei's high-frequency drugs in the treatment of ASD for network pharmacology research.TCMSP and Swiss target prediction database were used to obtain the active components and targets of drugs.The targets of ASD were obtained by Genecard,OMIM,DisGeNET,TTD and DRUGBANK databases.Using Cytoscape 3.7.2 to construct the network diagram and determine the core targets of drugs in the treatment of diseases.And to analyze the Go(gene ontology)function and KEGG(Kyoto Encyclopedia of genes and genomes)pathway enrichment of key targets by using Metascape database.Research result:1.A total of 125 children were included,including 105 males(84%)of the total,and 20 females(16%).The ratio of male to female is about 5.25:1.2.A total of 360 prescriptions and 209 traditional herbal medicines.The most frequent drug was Alpinia oxyphyllae fructus,followed by Acorus tatarinowii rhizoma.The four Qi of medicine are mainly cold and warm.The five flavor is mainly sweet,followed by pungent and bitter.The mainly channels were heart meridian and liver meridian.The most frequent drug efficacy was tonifying deficiency.The most frequent drug pairs were Alpinia oxyphyllae fructus and Acorus tatarinowii rhizoma.The drug combinations with higher association rules were:?Alpinia oxyphyllae fructus,Polygalae radix->Acorus tatarinowii rhizome;?Alpinia oxyphyllae fructus,Lilii Bulbus,Polygalae radix->Acorus tatarinowii rhizome;?Lilii Bulbus,Polygalae radix->Acorus tatarinowii rhizome.Set the support degree as 144 and the confidence degree as 0.8,the core prescriptions obtained from the network topology of traditional Chinese medicine are:Alpinia oxyphyllae fructus,Acorus tatarinowii rhizome,Uncariae ramulus cum uncis,Lilii Bulbus,Polygalae radix,Rehmanniae radix praeparata,Citri reticulatae pericarpium,Aconiti lateralis radix praeparata,Testudinis carapax et plastrum,Bambusae caulis in taenias,Curcumae radix,Moutan cortex.Based on K-means algorithm,six drug combinations were obtained.3.In this study,eight important active components including Stigmasterol,kaempferol and Cycloartenol were obtained from Alpinia oxyphylla fructus and Acorus tatarinowii rhizoma.There are 126 effective targets for the treating ASD,including AKT1,MAPK3,APP,EGFR,VEGFA,etc.Go enrichment analysis showed that the drug pair was mainly involved in the biological function of synaptic transmission.KEGG pathway enrichment analysis showed that neuroactive ligand receptor interaction,pathways in cancer,calcium signaling pathway were the main pathways of the drug pair.Research Conclusion:1.Professor Wang Sumei thinks that ASD in children is located in the brain,and its pathogenesis is disharmony of viscera,disorder of Qi,obstruction of pathogenic factors and disturbance of gods.Therefore,cold and warm drugs are often used for treatment.It is necessary to use the product of replenishing intelligence,opening orifices and calming the nerves.It emphasizes the movement of Qi mechanism and the harmony of Yin and Yang.It is usually used for regulating the heart and soothing the liver.At the same time,it is combined with tonifying,regulating qi and resolving phlegm drugs according to syndrome differentiation,so as to make Qi mechanism smooth,clear orifices nourish and mind gradually normal.2.The effect of Alpinia oxyphyllae fructus and Acorus tatarinowii rhizome in the treatment of ASD embodies the characteristics of multi-component,multi-target and multi-channel of traditional Chinese medicine.Its mechanism of action may be that the drug acts on the synapse and exerts anti-inflammatory,anti-cancer and anti-oxidation effects through multiple signaling pathways such as neuroactive ligand receptor interaction,cancer pathway and calcium ion signaling pathway.
