To Explore The Medication Rules Of Traditional Chinese Medicine In The Treatment Of Triple-negative Breast Cancer Based On Data Mining

BACKGROUNDIn recent years,the incidence of breast cancer has shown an obvious upward trend.According to the statistics of global malignant tumors in 2020,the incidence of breast cancer has ranked first among malignant tumors,and triple negative breast cancer(TNBC)accounts for about 10%-20%of all types of breast cancer.TNBC is a special subtype with different clinicopathological and biological characteristics from other breast cancer subtypes.It has the characteristics of early onset age,high recurrence rate,high metastasis rate and poor prognosis,which is one of the hot and difficult points in current research.Modern medical research found that endocrine therapy and anti-HER-2 molecular targeted therapy are not suitable for this subgroup.However,based on the particularity of this subgroup and the limitations of treatment methods,targeted therapy,immunotherapy and anti-androgen therapy have achieved certain results,but the applicable population is limited and further research is needed.Therefore,the main treatment methods are still surgery,chemotherapy and radiotherapy.Radiotherapy and chemotherapy can easily cause adverse reactions such as decreased immune function,bone marrow suppression,nausea and vomiting,alopecia,etc.,and reduce patients' compliance,so it is difficult to complete the treatment plan in sequence;Or produce resistance to chemotherapy drugs,leading to the failure of the treatment plan.In view of the high cost of western medicine treatment,drug resistance and adverse reactions,a good solution has not yet been found.Therefore,it is an urgent problem to seek drugs with low price,which can reverse drug resistance,reduce side effects,control metastasis,reduce tumor recurrence and improve the quality of life and prolong life of tumor patients.Tracing back to the ancient books,Chinese medicine believes that "qi deficiency is a rock","healthy qi exists inside,evil cannot be done",and deficiency of healthy qi is the root cause of cancer.Traditional Chinese medicine has a long history of understanding and treating breast cancer,and has accumulated a lot of theoretical knowledge and practical experience.With the development of modern technology,pharmacological research,animal experiments and clinical observation have fully proved that Chinese medicine can not only inhibit the growth and metastasis of TNBC,but also reverse drug resistance,improve the sensitivity of radiotherapy and chemotherapy,reduce toxic and side effects,thus enhancing patients'compliance and helping to successfully complete Western medicine treatment programs,which has achieved good results in clinic and is worth inheriting and learning.In view of the fact that there is no unified treatment guide for TNBC with traditional Chinese medicine as a guide,many doctors are flexible in clinical medication and have different compatibility rules.In recent years,more and more related literatures have been published,which will be helpful for clinical reference.OBJECTIVEThe purpose of this study is to explore the commonly used drugs in the treatment of TNBC,the compatibility and association rules between drugs and drugs,etc.,and to summarize the rules of drug use,in order to provide clinical diagnosis and treatment ideas and data support,and provide ideas for basic research.METHODSThis study retrieved and collected clinical literature on the treatment of TNBC by the oral administration of traditional Chinese medicines published in CNKI,Weipu Chinese Science and Technology Full-text Database,and Wanfang Chinese Database from January 1991 to February 2021.According to the inclusion criteria and exclusion criteria,140 prescriptions were selected,and the data were entered into EXCEL 2016 software,With reference to Pharmacopoeia of the People's Republic of China(2020 edition),Materia Medica of China and Chinese Pharmacy,the second edition of the national planning textbook for Chinese medicine colleges and universities,the names of Chinese medicines entered in the literature are standardized and consistent,and a database is established;Then,SPSS Modeler 18.0 and SPSS Statistics 20.0 are used to make statistics and analysis on the frequency,association rules and clustering results of the data,so as to mine the medication rule.RESULTS1 Literature publication year statisticsIn the clinical literature on the treatment of TNBC with traditional Chinese medicine,a total of 103 articles were included.Among them,0 qualified documents were published in 1991-2008,1 article in 2009,5 articles in 2010,1 article in 2011,4 articles in 2012,4 articles in 2013,5 articles in 2014,6 articles in 2015,17 articles in 2016,4 articles in 2017,20 articles in 2018,19 articles in 2019,16 articles in 2020,and 1 article in 2021.Since 2009,the statistics have been based on a 4-year interval,and a total of 11 articles were published in 2009-2012,a total of 32 articles were published in 2013-2016,and a total of 59 articles were published in 2017-2020.2 Medication frequency statisticsThere are 140 first prescriptions and 231 traditional Chinese medicines,with a total of 2133 times.Among the 30 traditional Chinese medicines with frequency over 20 times,the top 15 are Atractylodes,Bupleurum,Astragalus,Poria,White Peony,Licorice,Oldenlandia diffusa,Angelica,Codonopsis,Curcuma,Shansi Mushroom,Tangerine Peel,Chinese Yam,Turmeric,Chinese Wolfberry.According to the classification frequency of traditional Chinese medicine efficacy,the top three drugs are tonic drugs(680 times)31.88%,heat-clearing drugs(383 times)17.96%,and blood-activating and stasis-removing drugs(263 times)12.33%.3 Statistics of meridian tropismThe first two of the four qi:warm 729 times,cold 713 times;The top three flavors:sweet taste up to 1143 times,bitter taste 1077 times and pungent taste 804 times;The first five meridians:liver meridian 1141 times,spleen meridian 994 times,lung meridian 857 times,stomach meridian 634 times and kidney meridian 564 times.4 Association rule statisticsSetting the support degree to 20 and the confidence degree to 80%,a total of 20 results were derived.Among them,according to the frequency of drug combinations from high to low,the first five items are ?"Astragalus,Atractylodes",?"Poria Atractylodes macrocephala",?"White peony,Bupleurum",?"Curcuma,Atractylodes",?"White peony,Atractylodes,Bupleurum";sorted in descending order of confidence,The first five are?"Ligustrum lucidum,Atractylodes lanceolata",?"Poria,Astragalus,Atractylodes lanceolata",?"Hedyotis diffusa,Astragalus,Atractylodes lanceolata",?"White peony root,Astragalus,Atractylodes lanceolata",?"Ligustrum lucidum,Astragalus".Association rules are based on confidence ranking.5 Cluster analysisAccording to the tree cluster diagram,the first 25 traditional Chinese medicines were classified into three categories.The first category included Bupleurum,White Peony,Angelica,Tangerine Peel,Pinellia,Shanzi Mushroom,Zhejiang Fritillaria,Coix Seed,Taizi Ginseng,Poria,Licorice;The second category includes Dogwood,Rehmannia glutinosa,Yam;The third category includes Curcuma,Epimedium,Codonopsis,Oldenlandia diffusa,Turmeric,Prunella vulgaris,Scutellaria barbata,Lycium barbarum,Ligustrum lucidum,Atractylodes,Astragalus.CONCLUSIONThe human body is a complex organic whole,and the pathogenesis of TNBC patients is complicated.The high-frequency medication,drug combination,and prescription classification rules obtained through data mining must be combined with the patient's symptoms,tongue,pulse,etc.to be flexible in dialectics.Medication.
