Experimental Study Of Yishen Jiangu Granules On Osteoclasts By Regulating OPG/RANKL/RANK Pathway Via BMSCs

OBJECTIVE: To observe the experimental study of Yishen Jiangu Granules on osteoclasts via BMSCs-regulated OPG/RANKL/RANK pathway,and to provide important experimental references for Yishen Jiangu Granules to treat PMO.Methods: Experimental rats were deprived of bilateral ovaries to create models,which were divided into experimental group(castration + Yishen Jiangu granule gavage),model group(castration + normal saline gavage),and sham operation group(sham operation + normal saline gavage).Stomach),positive drug control group(castration +alendronate sodium gavage).After successful modeling,BMSCs were extracted,and BMSCs and bone marrow mononuclear cells were co-cultured in the upper and lower chambers of the Transwell chamber.The upper chamber was divided into: experimental group + osteoclasts,model group + osteoclasts,sham operation group + Osteoclasts,positive drug group + osteoclasts.Add osteoclast inducer to the lower chamber,observe the differentiation and maturity of osteoclasts by inverted phase-contrast microscope and count them,and detect Wnt10 b,?-Catenin,OPG and RANKL in BMSCs by realtime fluorescent quantitative PCR and Western blot.Results: 1.Yishen Jiangu Granules can increase the bone density of osteoporotic rats,and its effect is the same as that of alendronic acid.2.In the co-culture system,the number of osteoclasts model group+OC>positive control+OC group>experiment+OC group>sham operation+OC group.3 In the co-culture system,Wnt10 b,?-catenin,OPGm RNA and protein table in BMSCs,the model group + OC had the lowest expression,and the sham operation + OC group had the highest expression.After treatment with Yishen Jiangu granules and alendronic acid,Wnt10 b can be increased.,?-catenin,OPGm RNA and protein expression,and the effect of Yishen Jiangu Granules is better than that of alendronic acid.The expression of RANKL m RNA and protein in BMSCs was the highest in the model group + OC,and the lowest in the sham operation+ OC group.After treatment with Yishen Jiangu Granules and alendronic acid,the expression of RANKL and protein can be reduced,and the effect of Yishen Jiangu Granules Better than alendronic acid conclusion:1 Yishen Jiangu Granules can increase the bone density of osteoporotic rats,and its effect is the same as that of alendronic acid.2 Yishen Jiangu Granules can regulate the expression of OPG and RANKL in BMSCs cells,activate the OPG/RANKL/RANK signaling pathway to inhibit the differentiation and maturation of osteoclasts,and the Wnt/?-catenin signaling pathway may be involved in its regulation.
