Objective:Lipopolysaccharide(LPS)solution was injected intraperitoneally to construct a rat model of ALI/ARDS caused by sepsis,and then three different doses of tanshinone ?A were intervened with high,medium and low doses to pass the general behavioral evaluation of rats,Pathological blood,platelet count,APTT,PT,serum detection of IL-1?,IL-6,TNF-? and alveolar lavage fluid detection of t-PA,PAI-1levels,superalbumin content,and WB detection of lung tissue uniformity The expression level of thrombomodulin TM in the plasma,to explore the intervention effect of Tanshinone ?A on ALI/ARDS rats with sepsis.Methods:50 SPF male rats were randomly divided into 5 groups.The sham operation group(group A)was intraperitoneally injected with the same amount of 0.9% NS,and the other groups were intraperitoneally injected with 7mg/kg LPS,including high,medium and low doses of Tanshinone ?A One hour after the injection of lipopolysaccharide,the group was intraperitoneally injected with tanshinone ?A 80mg/kg,40 mg/kg,20 mg/kg,and all rats were sacrificed 24 hours later.Arterial blood was collected from the abdominal aorta,centrifuged to collect serum,ELLSA was used to detect IL-1?,IL-6,and TNF-? levels;venous blood was collected from the inferior vena cava to detect the number of platelets,APTT,and PT;the alveolar lavage fluid was collected,ELLSA was used to detect the expression levels of t-PA and PAI-1;the left lung was ligated at the same time when the alveolar lavage fluid was collected,the upper lobe of the left lung was taken to measure the dry-wet ratio,and the lower lobe of the left lung was taken for pathological sections.The right lung was used for lung tissue homogenization and Western Blot was used to detect the expression level of thrombomodulin.Results:1.Comparison of pathological evaluation of each group: The pathological score of the model group is high,and the pathological score of the low-dose group is relatively low,which is significantly different from group A(P<0.05);2.Comparison of pulmonary edema: the alveolar lavage superalbumin and W/D ratio of the model group and the high-dose group are high,and the pulmonary edema is severe.The difference between the two is not significant(P>0.05),but compared with other groups There is a significant difference in comparison(P<0.05);among them,the low-dose group has the lowest value after group A,but the difference is not significant(P>0.05);3.Comparison of platelets and APTT and PT: The platelets of group B and group C were significantly reduced,which was significantly different from other groups(P<0.05);compared with other groups,the model group had the shortest APTT and PT(P<0.05);The high-dose group had the longest APTT and PT(P<0.05);while the low-dose group had the longest APTT,PT,and thrombocytopenia,but they were the closest to the normal group(P>0.05);4.Comparison of alveolar lavage fluid PAI-1 and t PA: PAI-1 and t PA in group B are both higher,but the increase in PAI-1 is more obvious,and the imbalance of PAI-1/t PA is different from other groups(P< 0.05);Group E has the lowest increase in PAI-1,which is significantly different from group B and group C(P<0.05);5.Serum IL-1?,IL-6,and TNF-? levels: from high to low respectively;group B,group C,group D,group E,among which there is a difference between group DE and group B(P<0.05);group E has the best effect Good,there is no significant difference between the high-dose group and the model group(P>0.05);6.Comparison of lung tissue homogenate TM: Group B has the highest TM expression level and severe endothelial damage,which is different from other groups(P<0.05);the expression level of E group is the closest to that of group A,and is significantly different from group B(P<0.05);Conclusion:1.Sepsis can cause acute lung injury;intraperitoneal injection of LPS in rats can cause sepsis ALI/ARDS model.2.Sodium tanshinone ?A sulfonate plays a therapeutic effect on acute lung injury by reducing pulmonary edema,inflammation levels,vascular endothelial damage,regulating blood coagulation and reducing pulmonary microthrombosis.3.Sodium tanshinone ?A sulfonate has an interventional effect on acute lung injury,but its effect is related to the dose.Among them,the low-dose has the best effect,while the high-dose and middle-dose groups have poor effects.The high-dose group may generally have the same effect as early hypocoagulation.Leading to bleeding is not conducive to limiting inflammation in the local area.
