Objective: 1.To study whether tangerine peel and its effective parts have protective effects on lung injury in smoking rats.2.To study the effects of tangerine peel and its effective parts on the expression of IL-1?,IL-8,TNF-?,SOD,MDA,GSH-PX in the lungs of smoking rats.Methods: 80 SPF male rats were randomly divided into 10 groups by number method.The lung injury model of rats was made by smoking method.Rats in each experimental group were given tangerine peel preparation by gavage before each smoking,group A and B were given etc.Gavage the stomach with distilled water.The rats were sacrificed on the31 st day,and their alveolar lavage fluid and lung tissue were collected for the next study:(1)Observe the airway of rats by using the HE and MASSON staining methods on pathological sections of rat lung tissue specimens And the pathological conditions of alveolar inflammation and fibrosis to determine whether the modeling is successful,whether there are typical airway inflammation and alveolar fibrosis changes,and the influence of tangerine peel and its effective part intervention;(2)ELISA method detects BALF,IL-1?,The level of IL-8 and TNF-?.(3)The kit detects the content of MDA,SOD,and GSH-PX in lung tissue homogenate.Results: The alveolitis score of group B was significantly higher than that of group A,and the difference was significant(P<0.01).Compared with group A,the pulmonary fibrosis score of group B was significantly improved,and the difference was significant(P<0.01).The scores of alveolitis and pulmonary fibrosis of rats in each experimental group were not significantly different from that of group B,and there was no difference(P>0.05).The levels of IL-8,IL-1?,and TNF-? in group B were significantly higher than those in group A,and the difference was significant(P < 0.01).The levels of IL-8 in groups C,D,and E were significantly lower than those in group B,and the difference was significant(P<0.01).The levels of IL-8 in groups F and I were lower than those in group B,and there was a difference(P < 0.05).The levels of IL-1? in groups C and D,groups E and F were significantly lower than those in group B,and the difference was significant(P<0.01).The level of IL-1? in group I was lower than that in group B,which was different(P<0.05).The level of TNF-? in groups C,D,F,I,and J was lower than that in group B,which was different(P<0.05).The level of TNF-? in group E was lower than that in group B,and the difference was significant(P<0.01).The level of MDA in group B was significantly higher than that of group A,and the levels of SOD and GSH-PX in group B were significantly lower than those of group A,with significant difference(P<0.01).The levels of MDA in groups C,D,E,F,and I were significantly lower than those in group B,and the difference was significant(P<0.01).The level of MDA in groups G,H,and J was lower than that in group B,which was different(P<0.05).The activity of SOD in groups C,D,E,F,I was significantly higher than that in group B,and the difference was significant(P<0.01).The activity of SOD in group G was higher than that in group B,which was different(P<0.05).The activity of GSH-PX in groups C,D,E,F,I,and J was significantly higher than that of group B,and the difference was significant(P<0.01).The activity of GSH-PX in group G was higher than that in group B,there was a difference(P<0.05).Conclusion: 1.The method of smoking through the self-made smoking and poisoning box can successfully create a model of lung injury rats.2.Groups C,D,E,F,G,J,I,and J have no significant improvement in the pathological results of lung injury caused by smoking.3.C,D,E,F can reduce the levels of IL-8,IL-1?,TBF-?,MDA in lung tissue,and can increase the activity of SOD,GSH-PX,indicating that tangerine peel decoction and volatile oil extraction The substance may protect the lung damage caused by smoking by reducing the infiltration and damage of inflammatory cells in the lung tissue and regulating the oxidation/antioxidation balance in the lung tissue.4.Groups G and H can reduce the level of MDA in lung tissue,and have no effect on IL-8,IL-1?,and TNF-?.Group G increases the activity of SOD and GSH-PX,indicating that tangerine peel alkaloid extracts It may be to protect the lung damage caused by smoking by adjusting the balance of oxidation/antioxidation in the lung tissue,and relatively speaking,high doses are more effective.5.Groups I and J can increase the activity of GSH-PX,reduce the levels of TNF-?,MDA,group I can reduce the levels of IL-8,IL-1?,and increase the activity of SOD,indicating the possibility of tangerine peel flavonoids extract It protects the lung injury caused by smoking by inhibiting the inflammatory response in lung injury and adjusting the balance of oxidation/antioxidation.Relatively speaking,the high-dose group is better.
