Objective:To determine the chemical components of Tangerine Peel and Bamboo shavings Decoction,and to screen the active components through network pharmacology.Objective to establish the content determination method and fingerprint analysis method of multiple index components of Tangerine Peel and Bamboo shavings Decoction,and finally realize the overall quality control of Tangerine Peel and Bamboo shavings Decoction.Method:1.Chemical composition analysis of Tangerine Peel and Bamboo shavings Decoction:ultra high performance liquid chromatography quadrupole time of flight mass spectrometry(UPLC-Q-TOF/MS)was used for on-line separation and analysis of Tangerine Peel and Bamboo shavings Decoction samples.One dimensional retention time of UPLC was used to lock compounds.The molecular formula was determined by two-dimensional accurate mass number and isosteric kurtosis ratio The structural formula is deduced.2.The network pharmacology research of Tangerine Peel and Bamboo shavings Decoction:through tcmsp database,the active components of the decoction were screened,and the active components were uploaded to the pharmapper server to obtain the target protein of the active components.The genes of related diseases were retrieved from DrugBank,GeneCards,DisGeNET and OMIM databases.The network of traditional Chinese medicine active components target is constructed by using cyberscape software.The go enrichment analysis and KEGG pathway annotation analysis are carried out through the biological information annotation database(DAVID).3.Study on the quality standard of Tangerine Peel and Bamboo shavings Decoction:UPLC method was used with Waters Acquity UPLC HSS T3 C18(100×2.1mm,1.8μm)column;0.1%formicacid-acetonitrile as mobile phase.The fingerprints of 15 batches of Tangerine Peel and Bamboo shavings Decoction were established,and the similarity of 15 batches of Tangerine Peel and Bamboo shavings Decoction was evaluated by"similarity evaluation system of chromatographic fingerprints of traditional Chinese medicine".At the same time,the content of the index components in Tangerine Peel and Bamboo shavings Decoction was determined by HPLC.Result:1.Chemical composition analysis of Tangerine Peel and Bamboo shavings Decoction:the structure of 138 compounds in the decoction was identified or inferred.Among them,there are 59 flavonoids,55 triterpenoids,12 gingerols,3 limonoids and 9 other components.Flavonoids mainly come from tangerine peel and licorice,triterpenoids mainly come from licorice and ginseng,gingerols mainly come from ginger.2.Study on network pharmacology of Tangerine Peel and Bamboo shavings Decoction:24 potential active components were obtained from the chemical constituents of Tangerine Peel and Bamboo shavings Decoction.620 component targets were uploaded to pharmmappee server,and 6569disease targets and 404 intersection targets were obtained by DrugBank、GeneCards、DisGeNET and OMIM database with acute and chronic gastritis,gastroesophageal reflux and pyloric obstruction as key words.Cytoscape software was used to analyze the component target network,and 10 active components were obtained.According to P<0.05,441 biological processes,110 molecular functions and 60cell components were obtained by go analysis,and 111 signal pathways were obtained by KEGG analysis.3.Study on the quality standard of Tangerine Peel and Bamboo shavings Decoction:five methods for the determination of the content of the components of the indexes were established,including hesperidin and rutingin from the aged skin,glycyrrhizin and ammonium glycyrrhizate of Glycyrrhiza and ginsenoside Rb1 of ginseng.The fingerprint analysis method of Tangerine Peel and Bamboo shavings Decoction was established.12 common characteristic peaks were determined by 15 batches of freeze-dried powder of Tangerine Peel and Bamboo shavings Decoction,and the origin of the medicinal materials was assigned,and 7 chromatographic peaks were identified.Conclusion:UPLC-Q-TOF/MS technology can quickly and accurately identify the chemical components in the Tangerine Peel and Bamboo shavings Decoction,and lay the foundation for its network pharmacology research;through the network pharmacology,the main active components of the decoction are predicted,which provides the basis for the establishment of the index components in the quality standard research;the HPLC multi-index component content determination of the angerine Peel and Bamboo shavings Decoction is practical The UPLC fingerprint of angerine Peel and Bamboo shavings Decoction,from the index component to the quality control of the medicinal materials,directly reflects the differences between different batches.The combination of the content determination of multiple components and fingerprint provides a reference for its quality evaluation.
