Objective: This subject research explores the effect and mechanism of Xieqing Pills on TS model rats from the perspective of neurotransmitter imbalance,and preliminarily explores the possible microscopic nature of TCM liver wind.Methods:Fifty rats(3 weeks old,SPF grade)were randomly divided into 5 groups:blank group,model group,tiapride group,high-dose Xieqing pill group,and low-dose Xieqing pill group,with 10 rats in each group.Except for the blank group,the other groups used IDPN to prepare TS rat models and conduct behavioral observations.After the successful modeling,they were sacrificed after two weeks of drug intervention.The head was decapitated,the brain was removed,and the striatum was removed.The product HVA is quantitatively detected by ELISA.Results:(1)The effect of Xieqing pill on the behavior of TS model rats: After intraperitoneal injection of IDPN,rats showed increased head vertical movement,rotation behavior and other stereotyped behaviors,as well as increased visitation behavior,overexcitement and other motor behavior changes,indicating that the model was successful.Compared with the model group,the Tiapride group had no significant difference in improving the behavior of rats.Traditional Chinese medicine intervention can improve the behavioral score of rats,but there is no significant difference between the two groups of Chinese medicine.In terms of weight change,with the intervention of drugs,compared with the model group,the weight of the blank group,the high-dose Xieqing pill group,and the low-dose Xieqing pill group all increased significantly(P<0.01).There was no significant difference between the two groups of traditional Chinese medicine..There was no statistically significant difference between the Tiapride group and the blank group,model group,and high-dose Xieqing Pill group.Compared with the Tiapride group in the low-dose Xieqing pill group,the rats in this group gained more weight(P<0.05).(2)The effect of Xieqing Pills on DA and HVA in the brain of the model group: Compared with the blank group,the dopamine content of the model group increased significantly,P<0.01,the difference was obviously statistically significant.Compared with the model group,the dopamine in the Tiapride group,the high-dose Xieqing Pill group,and the low-dose Xieqing Pill group decreased significantly,P<0.01,the difference was obviously statistically significant.However,the difference between the three groups of dopamine was not significant,P>0.05,which was not statistically significant.Compared with the blank group,the hypervanillic acid of the model group increased significantly,P<0.05,the difference was statistically significant.Compared with the model group,the dopamine metabolite hypervanillic acid of the Tiapride group,the high-dose group of Xieqing pills,and the low-dose group of Xieqing pills decreased significantly,P<0.01,there was no statistical difference between the three groups of high vanillic acid content.significance.Conclusion: Xieqing pills can improve the stereotyped behavior and abnormal motor behavior of TS model rats,and its mechanism of action may be related to the regulation of the metabolism of the monoamine neurotransmitter dopamine in the brain.The metabolic disorder of dopamine may be the internal microscopic nature of the imbalance of yin and yang in the liver and wind in traditional Chinese medicine.
