Objective:1.Through the intervention of magnetic field,to explore the pathological basis of magnetic field affecting blood stasis syndrome of coronary heart disease,and to provide new theoretical basis for physical intervention methods of blood stasis syndrome of coronary heart disease.2.By studying the effect of magnetic field on DES and CTNNB1 gene methylation expression in rats with coronary heart disease and blood stasis syndrome,the epigenetic regulation mechanism of magnetic field intervention on DNA methylation of coronary heart disease and blood stasis syndrome was expounded.Methods: The experimental animals were divided into three groups,the first is normal group and the rest is the model group.In the model group,depending on the presence or absence of magnetic field intervention,there were coronary heart disease with blood stasis syndrome group and coronary heart disease blood stasis syndrome with magnetic field intervention group.Electromagnetic field with magnetic field intensity of 0.5-12000 hz was used for magnetic field intervention,30 minutes a day for 14 days.After the intervention of magnetic field,the rats were anesthetized and fixed,and their blood lipids and hemorheology were detected.The heart and aorta tissues were stained with hematoxylin eosin,and their structural changes were observed.The DNA methylation status before and after magnetic field intervention was analyzed by control study method.Pyrosequencing method was used to analyze and study the coronary heart disease specific genes DES and CTNNB1 in the three groups.The reaction results of coronary heart disease blood stasis syndrome specific genes were obtained.Results: Pathological indexes: blood lipid: the value of CHO1,LDL-C and HDL-C in the intervention group of coronary heart disease and blood stasis syndrome group were significantly higher than that in the blank group(P < 0.05 or P < 0.01);There was no significant difference between the three groups(P > 0.05).Hemorheology: there was no difference in the viscosity of the three groups(P > 0.05);Compared with the blank group and the coronary heart disease blood stasis syndrome group,the values of low shear viscosity,medium shear viscosity and high shear viscosity were significantly increased(P < 0.05 or P < 0.01);The histopathological examination of heart and aorta showed that there was no significant difference between the group of coronary heart disease and the intervention group of magnetic field of coronary heart disease blood stasis syndrome,and the heart and aortic arch were all atherosclerosis in three stages.In the methylation analysis,the methylation level of Des gene was higher in the magnetic field intervention group than in the coronary heart disease blood stasis syndrome group,while compared with the blank group,the methylation level of CTNNB1 gene was higher than that of the blank group(P < 0.05).Conclusion:1.The model of coronary heart disease with blood stasis syndrome is successful,and the magnetic field intervention may not change the lipid metabolism disorder and improve the blood environment of coronary heart disease rats with blood stasis syndrome due to the influence of treatment time,amplitude and intensity;2.Magnetic field intervention can up regulate the methylation level of DES and CTNNB1 to a certain extent,so as to inhibit the gene expression,which has a certain adjuvant therapeutic effect on blood stasis syndrome of coronary heart disease.
