Objective:1.To explore the possible target and mechanism of Quyu Pinggan Huatan Decoction in the treatment of hypertension,and to provide support for animal experiments.2.To clarify the regulation effect of Quyu Pinggan Huatan Decoction on hypertension,to explore its intervention on blood pressure based on the RAAS pathway,and to explore its protective effect on vascular endothelium by detecting the expression of ET-1,to provide experimental basis for the treatment of hypertension.Methods:1.Using the pharmacological database of traditional Chinese medicine system and the analysis platform,the network diagram of"traditional Chinese medicine-component-target-pathway" of Quyu Pinggan Huatan Decoction was established to dig out the biological processes that may be related to the treatment of hypertension by Quyupinggan Huatan Decoction.2.Twenty SD rats were divided into 4 groups by using random number method in SPSS statistical analysis software,which were blank group,model group,traditional Chinese medicine group and western medicine group,with 5 rats in each group.Except the blank group,the remaining 3 groups were used to establish the hypertensive rat model by L-NNA(concentration:1mg/mL)at 15mg/(kg·d).After successful modeling,the blank group(3mL purified water/animal/day),model group(3mL purified water/animal/day),western medicine group(Captopril suspension 3mL/animal/day,about 0.605mg)and Chinese medicine group(concentrated crude medicine solution 3mL/animal/day,about 6.095g)were given continuous administration for 4 weeks;SBP and DBP of rats in each group were measured before intragastric administration and on the weekend morning of every weeks,1 hours after intragastric administration.Four groups of rats in the last 2 h after gastric drug materials,by the method of ELISA to detect serum Renin,Ang ?,the content of ALD section,HE staining to observe the thoracic aorta immunohistochemical detection of the content of ET-1 artery.Results:1.To remove phlegm stasis flat liver soup drugs,after the database mining found and treatment of hypertension related targets for 916,the final 23 key protein targets is obtained by screening,including IL6,NOS3,AKT1,VEGFA,propagated,REN,ACE,etc.,these protein targets involved in inflammatory signaling pathways,physiological processes of REDOX reaction,vasoconstriction,etc.2.The animal experiment results showed that the SBP and DBP of the blank group and the two groups were decreased compared with the model group at different time points after administration,the difference was statistically significant(P<0.01).He staining showed that the intimal structure of the aorta of rats in the traditional Chinese medicine group and the western medicine group was basically perfect,and the boundary between the arterial membranes was cl ear.ELIS A method to detect serum Renin,Ang ?,the expression of ALD,the blank group,the traditional Chinese medicine,western medicine group after 4 weeks to fill the stomach,Renin,Ang?,low ALD are expressed in the model group,the difference is statistically significant(P<0.01);Lavage compared two groups,Renin,Ang ?,ALD index differences,does not have statistical significance(P>0.05);Microscopic observation of the immunohistochemical results showed that the expression of ET-1 in the model group was significantly higher than that in the blank group(P<0.01)and the drug administration groups(P<0.05),the differences were statistically significant,but there was no significant difference between the two groups(P>0.05).Conclusion:1.Quyu Pinggan Huatan Decoction can intervene blood pressure through multiple channels and targets,and can play a role of comprehensive regulation and antihypertensive effect.2.Removing phlegm stasis flat liver soup can intervene LNNA induced hypertension rats blood pressure,and can reduce the Renin,Ang ?,the expression of ALD,by regulating the RAAS,at the same time can reduce the expression of intravascular ET-1,have a role to protect the vascular endothelium.
