Objective: To observe the effect of xuanbitongluo ointment on the swelling degree of ankle joint,pathological changes of joint and Notch1 level in serum and joints of rats,and to analyze the mechanism of xuanbitongluo ointment on acute gouty arthritis model rats.Methods: 30 Wistar male healthy rats were randomly divided into 6 groups:xuanbitongluo ointment high dose group,xuanbitongluo ointment medium dose group,xuanbitongluo ointment low dose group,colchicine group,model group and blank group,with 5 rats in each group.After recording the initial perimeter of the right ankle joint,the model was established.MSU solution was injected into the right ankle joint of rats in xuanbitongluo ointment high,medium and low groups,colchicine group and model group,and the same volume of normal saline was injected into the blank group.After successful modeling,the rats in each group began to gavage and recorded the ankle circumference 24 h,48 h and 72 h after modeling.After the last measurement of the joint length,the blood samples were taken from the abdominal aorta,and the concentration of Notch1 in serum was detected by ELISA.After death,the right ankle tissue was cut,and the pathological observation was performed by HE staining,and the expression of Notch1 in the samples was detected by Western blot Statistical analysis was performed after the treatment.Results: 1.48 hours after modeling,the swelling degree of high-dose xuanbitongluo ointment group was significantly lower than that of model group(P < 0.05),the difference was statistically significant;72 hours after modeling,the swelling degree of high-dose,medium dose and low-dose xuanbitongluo ointment groups was significantly lower than that of model group(P < 0.05),the difference was statistically significant.2.The degree of articular cartilage injury and tissue proliferation in high,medium and low dose xuanbitongluo ointment groups was significantly lower than that in model group.3.The serum Notch1 level of xuanbitongluo ointment high-dose group and middle dose group was significantly lower than that of model group(P <0.05),the difference was statistically significant;there was no difference between xuanbitongluo ointment low-dose group and model group(P >0.05).4.The expression level of Notch1 in the high-dose xuanbitongluo ointment group was significantly lower than that in the model group and colchicine group(P < 0.05),the difference was statistically significant;the middle dose xuanbitongluo ointment group was significantly lower than that in the low-dose xuanbitongluo ointment group(P < 0.05),the difference was statistically significant.Conclusion: Xuanbitongluo ointment can reduce the level of Notch1 receptor protein in serum and joint tissue,improve the symptoms of acute gouty arthritis and the degree of articular cartilage damage.
