Effects Of Chaihu Sanshen Capsules On The Expression Of Bcl-2 And Fas In Myocardial Cells Of Ischemic Arrhythmia Rats

Objective: To observe the expression of Bcl-2 and Fas protein in myocardial cells of rats with ischemic arrhythmia treated by Chaihu Sanshen Capsule,and to understand the possible mechanism of Chaihu Sanshen Capsule in treating ischemic arrhythmia,so as to provide a new experimental basis for it.Methods: Methods: 60 SPF Wistar male rats were randomly divided into 6 groups: sham operation group,model group,Chaihu Sanshen Capsule group,Shensong Yangxin Capsule group,Verapamil group and blank group,10 rats in each group.After continuous feeding for 2w in each group,the ischemic arrhythmia rat model was prepared by ligation of coronary artery with silicone tube.The ecg changes during the whole modeling process were observed in 6 groups of rats,and the incidence of ventricular presystolic(VPB),ventricular tachycardia(VT)and ventricular fibrillation(VF)in each group were analyzed,and arrhythmia score was performed.The protein expression of bcl-2 and Fas in ischemic myocardium of rats was determined by Western blot.Results:1.Compared with the blank group,the incidence of arrhythmia in the sham-operated group increased slightly,but there was no significant difference(P > 0.05);the incidence of arrhythmia in the model group increased significantly,and the score of arrhythmia in the model group was significantly higher than that in the sham-operated group;the experimental model of ischemic arrhythmia was verified to be reliable and successful.Compared with the model group,the arrhythmia scores of Chaihu Sanshen capsule group,Shensong Yangxin capsule group and verapamil group decreased significantly(P<0.05).There was no significant difference between the Bupleurum Sanshen Capsule group and the Shensong Yangxin Capsule group(P > 0.05);there was significant difference between the Bupleurum Sanshen Capsule group and the Verapamil group(P < 0.05).2.Compared with the blank group,the expression level of Bcl-2 protein in myocardial cells of sham-operated group had no significant difference(P > 0.05).Compared with the model group,the expression level of Bcl-2 protein in myocardial cells of model group was relatively low,and that of Bcl-2 protein in verapamil group was relatively high.Both Chaihu Sanshen capsule group and Shensong Yangxin capsule group could increase the heart rate.The expression of myocyte apoptotic protein Bcl-2 was significantly different from that of drug intervention(P < 0.01).There was no significant difference in P value among the three groups of Chaihu Sanshen capsule group,Shensong Yangxin capsule group and verapamil group.3.Compared with the model group,the expression of Fas protein in the Chaihu Sanshen Capsule group,the Shensong Yangxin Capsule group and the Verapamil group decreased to a certain extent,and the difference was statistically significant(P<0.05).Bcl-2 decreased and Fas protein increased in the model group,indicating that myocardial apoptosis can occur after ischemic arrhythmia.Pre-intervention in the three groups of Chaihu Sanshen Capsule group,Shensong Yangxin Capsule group and Verapamil group could up-regulate the expression of Bcl-2 protein and down-regulate the Fas protein,all of which could resist the myocardial apoptosis caused by ischemic arrhythmia,and the effects of the three groups on reducing myocardial injury were not significantly different.Conclusion:1.Chaihu Sanshen Capsule can reduce the occurrence of ischemic arrhythmia.2.The expression of Bcl-2 was up-regulated in Chaihu Sanshen Capsule,Shensong Yangxin Capsule and Verapamil.3.The expressions of Fas in Chaihu Sanshen Capsule,Shensong Yangxin Capsule and Verapamil could be down-regulated.4.Chaihu Sanshen Capsule up-regulated the expression of inhibiting apoptotic factor Bcl-2 and down-regulated the expression of promoting apoptotic factor Fas,indicating that it could alleviate apoptosis and myocardial injury,providing experimental basis for Chaihu Sanshen Capsule to improve ischemic arrhythmia.
