Objective:Meridian-organism related is one of the important contents of acupuncture theory and has always been a research hotspot in the field of traditional Chinese medicine,and guides clinical diagnosis and treatment.Previous studies have focused on the repairing effect of acupuncture and moxibustion on pathological changes in organs,and the changes in the response of related molecular pathways or molecular substances to determine the efficacy of acupuncture and the mechanisms regulating the status of organs.Or when the viscera is in a normal physiological state,the influence of acupuncture on the anatomical or molecular substances in the plasma of the body's central or surrounding body.However,when the organs are in the normal physiological state or pathological state,there are very few reflections and basic researches on the surface meridians and acupoints.High-frequency currents have a certain regulatory effect on cardiovascular diseases,especially in myocardial ischemic injury.Previous studies have shown that the effects of high-frequency electrocardiography on the heart are mainly applied to basic experimental research,and the central and peripheral nerves have a certain regulatory effect on the heart.The research content mainly focuses on changes in heart-related effect indicators or molecular indicators after high-frequency electroacupuncture,which in turn produces a protective effect on the heart.However,when the myocardium is in different states(physiological,pathological,high-frequency electroacupuncture acupoints or non-acupoint intervention state),there are few studies on the surface meridians and acupoints and their molecular substances.In this study,a rat model of myocardial ischemia was used,and various modern techniques such as biophysics and molecular biology were used to observe the regularity of the skin temperature changes in the same meridians and different acupoints and the Changes and regularities of NO and CaM responses to sympathetic nerve-related molecules in skin tissue when the myocardium was in different states(normal physiological state,ischemic injury state,and electro-acupuncture intervention state).We try to found the important approach to explain the scientific connotation of the acupoint viscera correlation form the perspective of sympathetic correlates of biophysical phenomena and receptor molecular response changes.Methods:80 male Wistar rats were randomly divided into the electro-acupuncture one times group and electro-acupuncture three times group,and each group was divided into control group,sham-operation group,the model group,the high-frequency electro acupoint group and high-frequency electro nonacupoint group.Each group contained 8 rats.Both therapeutic groups were applied needle to "Neiguan"(PC6)after successfully producing the model of rats with myocardial ischemia.After the first or third time of EA treatment,the myocardial morphological changes were detected by echocardiography,and skin temperature was tested by infrared thermal imaging technology around "Neiguan"(PC 6)," Ximen(PC 4)”and" Tianquan(PC 2)".The content of NO by Nitrate reductive enzymatic to reflect NO contents around "Neiguan"(PC 6)and " Tianquan(PC 2)".The content of CaM around "Neiguan"(PC 6)and "Tianquan(PC 2)" were detected by ELIS A.Results:(1)The rat heart echocardiography:Compared with the normal group,the ejection fractions and left ventricular fractional shortening of model group were significantly decreased(P<0.01);Compared with the model group,the ejection fractions and left ventricular fractional of high frequency electro acupuncture group were significantly increased(P<0.01);the ejection fractions and left ventricular fractional of high frequency electro nonacupuncture group had no significant difference.(2)Change of skin tempereture:Longitudinal observation of changes in skin temperature in each group after one or three times treatment:compared with the normal group,the skin temperature of three acupounts in sham-operation group have no significant difference(P>0.05),and the model group were significantly increased(P<0.01);Compared with the model group,the skin temperature of three acupounts in high frequency electro acupuncture were significantly increased(P<0.01),the skin temperature of three acupounts in high frequency electro nonacupuncture had no significant difference(P>0.05).Lateral observation of changes in skin temperature in each group when transversing the same number of electro-acupuncture treatment:Compared with the model group,the changes in the temperature of the different cavernous areas were compared.The change of skin temperature in the "Neiguan" point area was larger than that of the two points,followed by the "Xianmen" point and finally the "Tianquan" point.Lateral observation of the difference in myocardial status in the same state,the same point area of treatment 1 and 3 times after the blank group and high frequency acupoint group compared with the model group,the difference after treatment is larger.(3)Content of NO:Longitudinal observation of changes in NO content in each group after one time treatment:compared with the normal group,the NO content of two acupounts in sham-operation group have no significant difference(P>0.05),and the model group were significantly increased(P<0.01);Compared with the model group,the NO content of two acupounts in high frequency electro acupuncture were significantly increased(P<0.01),the NO content of two acupounts in high frequency electro nonacupuncture had no significant difference(P>0.05).Longitudinal observation of changes of the content of NO in each group after three times treatment:compared with the normal group,the NO content of three acupounts in sham-operation group have no significant difference(P>0.05),and the model group were significantly increased(P<0.01);Compared with the model group,the NO content of"Niguan(PC 6)" acupount area in high frequency electro acupuncture were significantly increased(P<0.01),the NO content of "Tianquan(PC 2)" acupount area in high frequency electro acupuncture had no significant difference(P>0.05),the NO content of two acupounts in high frequency electro nonacupuncture had no significant difference(P>0.05).Lateral observation of the NO cntent changes in each group when transversing the same number of electro-acupuncture treatment:Compared with the changes in the temperature of different cavernous areas compared with the model group,the change of NO content in the"Neiguan" point area is relatively large.Lateral observation of the myocardium in the same state,the same point area after one or three times treatment,the difference between the NO content blank group and high-frequency acupoint group was compared with that of the model group.The "Neiguan" point area was higher after 3 treatments,and the "Tianquan" point area was higher.(4)Content of CaM:Longitudinal observation of changes in NO content in each group after one time treatment:compared with the normal group,the CaM content of two acupounts in sham-operation group have no significant difference(P>0.05),and the model group were significantly increased(P<0.01);Compared with the model group,the CaM content of two acupounts in high frequency electro acupuncture were significantly increased(P<0.01),the CaM content of two acupoints in high frequency electro nonacupuncture had no significant difference(P>0.05).Longitudinal observation of changes of the content of CaM in each group after three times treatment:compared with the normal group,the CaM content of two acupounts in sham-operation group have no significant difference(P>0.05),and the model group were significantly increased(P<0.01);Compared with the model group,the CaM content of"Niguan(PC 6)" acupount area in high frequency electro acupuncture were significantly increased(P<0.01),the CaM content of "Tianquan(PC 2)" acupoint area in high frequency electro acupuncture had no significant difference(P>0.05),the CaM content of two acupounts in high frequency electro nonacupuncture had no significant difference(P>0.05).Lateral observation of the CaM cntent changes in each group when transversing the same number of electro-acupuncture treatment:Compared with the model group,the change of CaM content in the "Neiguan" point area was relatively large.Lateral observation of the myocardium in the same state,the same point area after one or three times treatment,the difference between the CaM content blank group and the high-frequency acupoint group was compared with that of the model group.The "Neiguan"and "Tianquan" point areas were higher after 3 treatments.Conclusion:(1)In the evaluation of myocardial status,echocardiography showed that the activity of the anterior chamber wall in the model group decreased,suggesting myocardial ischemia in the left ventricular wall.The myocardial blood supply after high-frequency electroacupuncture treatment was improved to a certain extent.(2)In the skin temperature of the acupuncture point,the results of the three selected points indicated that there were certain increases in the "Neiguan(PC 6)","Ximen(PC 4)",and"Tianquan(PC 2)" points,reflecting the correspondence between the myocardium state and the pericardial meridians.(3)After high-frequency electroacupuncture treatment,the results of NO content in"Neiguan(PC 6)" and "Tianquan(PC 2)" points showed that the difference in NO content between different groups after electroacupuncture treatment indicated that different states of the myocardium were in "Neiguan(PC 6)","Tianquan(PC 2)" points are reflected on the points;while electro-acupuncture treatment is reflected on "Neiguan(PC 6)"points after 3 times,while "Tianquan(PC 2)" points are not specifically reflected,suggesting that there are relative specificity between different acupoints of the same meridian.(4)After high-frequency electroacupuncture treatment,the CaM content of skin tissue in the acupoint area showed that the difference between different groups after electroacupuncture treatment indicated that different states of the myocardium were found on the "Neiguan(PC 6)" and "Tianquan(PC 2)" points.It was reflected that electroacupuncture treatment was reflected on the "Neiguan(PC 6)" point after 3 times of treatment,while the "Tianquan(PC 2)"point was not specifically reflected,suggesting that there is relative specificity between different acupoints of the same meridian.
