Establishment diabetic mice model with underactive bladder and urodynamic evaluationObjectives Through the urodynamic study,we investigated the changes of bladder voiding dysfunction in diabetic mice induced by streptozotocin(STZ),in order to provide an experimental of animal platform for further clinical study of underactive bladder(UAB).Methods An animal model of diabetic mice was prepared by intraperitoneal injection of streptozotocin(STZ,65 mg/kg),the same age of mice as normal control group(n=10).Each group of mice body weight(BW)and fasting plasma glucose(FBG)were measured every two weeks,each group of mice bladder wet weight(BWW),24 hour urine volume(24hUV)and urodynamic examination were performed in twenty-fourth weeks after STZ induction,urodynamic parameters including intercontraction interval(ICI),maximum cystometric capacity(MCC),maximum bladder pressure during micturition(Pmax),void volume(VV),residual urine rate(RUR)and post-void residual urine volume(PVR).Results The 13 of 15 mice were successfully induced diabetic mice.Compared with the normal group,the FBG of diabetic mice with UAB was significantly increased,BW obviously decreased,24hUV and BWW significantly increased(P<0.05).The urodynamic parameters of diabetic mice with UAB showed that MCC,VV,RUR and PVR were significantly increased(P<0.05),ICI significantly prolonged and Pmax significantly decreased.Conclusions Intraperitoneal injection of STZ could establish a simple and effective model of diabetic mice,urodynamic evaluation were consistent with UAB changes in twenty-fourth weeks after STZ induction,and diabetic mice had similar micturition function changes with human.Therefore,the diabetic UAB model can be used as a reliable animal model for clinical research of UAB.The changes of bladder micturition function decompensation phase appeared during the late stage of diabetes mellitus,which might be associated with polyuria and hyperglycemia.Part? Improvement effect of sulforaphane on voiding dysfunction in diabetic mice with underactive bladderObjectives To investigate the improvement effect of antioxidant sulforaphane(SF)on bladder micturition function in diabetic mice with underactive bladder induced by streptozotocin(STZ).Methods An animal model of diabetic mice was prepared by intraperitoneal injection of streptozotocin(STZ,65 mg/kg).Successful diabetic mice were randomly divided into 3 groups:SF experimental group(intraperitoneal injection of SF,n=10),experimental control group(intraperitoneal injection of corn oil vehicle,n=10)and diabetic model group(n=10),the same age of mice as normal control group(n=10).Each group of mice body weight(BW)and fasting plasma glucose(FBG)were measured every two weeks,each group of mice bladder wet weight(BWW),24 hour urine volume(24hUV)and urodynamic examination were performed in twenty-fourth weeks after STZ induction,urodynamic parameters including intercontraction interval(ICI),maximum cystometric capacity(MCC),maximum bladder pressure during micturition(Pmax),void volume(VV),residual urine rate(RUR)and post-void residual urine volume(PVR).Results Compared with normal control group,3 groups of diabetic mice showed that BWW,24hUV,MCC,PVR and RUR were significantly increased,ICI significantly prolonged and Pmax significantly decreased.Compared with diabetic model group,experimental control group mice showed that BWW,24hUV,MCC,VV,PVR and RUR had no statistical difference(P>0.05).However,SF experimental group mice showed that BWW,24hUV,MCC,PVR and RUR were significantly decreased(P<0.05).Compared with experimental control group,SF experimental group mice showed that BWW,24hUV,MCC,PVR and RUR were significantly decreased(P<0.05),ICI shortened and Pmax slightly increased.Conclusions Antioxidant SF could reduce the damage degree of bladder voiding function in diabetic mice,and alleviate the symptoms of voiding dysfunction,which might be related to SF reduce oxidative stress on bladder detrusor tissue damage.
