Tristetraprolin(TTP)was a protein family that contains CCCH tandem zinc finger.The functions of TTP are binding mRNAs and transporting mRNAs into RNA degradation complex(P-body,Exosome)to degradation in animals.In Arabidopsis,the C3H14 and C3H15 are two homologous members of TTP family.C3H14 and C3H15 were expressed in anther microspore mother cell,and C3H15 expresses more strongly than C3H14.Microspore mother cell of these two gene double mutants(c3h14 and c3h15)were meiotically arrested in pachytene.The C3H15 are contained in P-body(Processing body)of anther.However,the molecular mechanism of TTPs regulating meiosis is not clear.In this study,the pull-down experiment verified the direct binding of C3H15 protein to DCP1/2,the core component of P-body.The RNA and protein affinity purification experiments proved that C3H15 is an RNA binding protein.Combined with sequencing and bioinformatics analysis,the binding genes,binding sites and motifs of C3H15 were obtained.After GO enrichment and expression profiling of C3H15-binding genes,These genes were found to be enriched in biological processes of in response to water loss,low temperature response,and cadmium ion response;cytoplasmic matrix of chloroplast-related cellular components;and molecular functions of protein and mRNA binding.Based on the expression analysis of the core components of P-body DCP1/2,it was revealed that P-body widely exists in different tissues and organs of Arabidopsis plants.These results are helpful for the composition and function analysis of P-body,as well as the in-depth study of the mechanism of C3H15 regulation of meiosis.
