MPF (maturation promoting factor), participate in the meiosis of oocytes, but the role and the control mechanism were not distincted. We intended to research the role of MPF during meiosis in male germ cells though our experiment, which would provide a reference to IVS (in vitro spermatogenesis) and optimize the condition of culture in vitro.In order to illuminate the expression regularity in different phases of spermatogenesis in mice, We aquird cell suspension from testes in distinct development phases by digesting and centrifuging. Then analyzed cell ploidy by Flow Cytometer, in addition analyzed the relationship among the phase feature of gonepoiesis, activity of MPF and gonepoiesis by immunohistochemical method combinding with the detection of activity. The result showed that in mice at the age of 8 days and 14 days there was no haploid, which appeared from 18d, but very few, when tetraploid increased to peaking, in addition the expression of the two subunit and the activity of MPF were at the high level. On the whole, in mice at the age of 18 days, most of spermatogenic cells were primary spermatocytes, the activity of MPF was the hightest and the result of development phases of spermatogenic cells coincided with the expression and activity result of MPF.For research the role of MPF in spermatogenesis, we discovered the best BL-Ⅰ(ButyrolactoneⅠ) concentration to inhibit activity of MPF is 0.68μmol/L, with the subject of 18 days mice, establishing coculture system and analyzing the activities of cells and MPF. At that concentration, BL-Ⅰcaused the rate decreasing of tetraloid and haploid, on the contrary the rate of diploid increased, which indicated MPF is essential to the second meiosis of spermatocyte.
