Objective: To study the differentiation process of white adipose tissue in rats with different degrees of obesity induced by high fat diet,and to explore the effect of subcutaneous adipose tissue differentiation in rats with different degrees of obesity after exercise intervention,so as to provide theoretical basis for exercise prevention and treatment of obesity and chronic diseases.Methods: In Experiment 180 rats were selected and divided into normal diet group(MC),super-recombinant group(ML),mild obesity group(MN)and moderate obesity group(MS)to establish an obesity model.After successful modeling,6 rats were sampled from each group and the indexes were detected.Experiment 2: The rats in the successfully modeled hyperreorganization,mild obesity group and moderate obesity group were divided into the quiet group(CL,CN,CS)and exercise group(TL,TN,TS),and the exercise group rats were trained on running platform intervention for 8 weeks.After the training,8 rats were sampled from each group for index detection.The body weight and fat weight of the rats were weighed,and the levels of IL-6,TNFA,Insulin and Leptin in the lipids of the rats were determined.After cell morphology was measured by HE staining,the number and volume of rat adipocytes were counted,and the protein expressions of BMP4,Smad1,Zn F423,PPARγ,FABP4 and LPL in subcutaneous adipose tissue were detected by Western-blot method,and then the data were statistically analyzed.Results: 1.Compared with MC group,each weighing index of rats in ML,MN and MS groups was significantly increased,and the ratio of seb-to-lipid and body fat were significantly increased.2.Compared with MC group,the higher the degree of obesity,the higher the levels of blood lipid(TC,TG,LDL-C),inflammatory cytokines,insulin and leptin.3.Compared with MC group,the higher the degree of obesity,the larger the volume of subcutaneous adipocytes,the less the number of adipocytes,and the lower the relative expression of adipodiation-related proteins.Experiment 2:1.Exercise significantly reduced the weighing indexes of obese rats,and decreased the sebum percentage and body fat percentage of rats.2.Exercise significantly reduced the levels of lipid(TC,TG,LDL-C),inflammatory cytokines,insulin and leptin in obese rats.3.Exercise significantly reduced the volume of adipocytes and increased the expression of differentiation-related proteins in obese rats.CONCLUSIONS: 1.In a certain degree of obesity,the expression of adipose differentiation protein in rats decreases with the increase of obesity.2.8 weeks of moderate-intensity exercise can significantly promote the increase of adipoprotein expression in rats with different obesity levels,and the benefit of exercise is not affected by obesity levels. |
