Abstract/Summary:
Cardiovascular diseases become major disease threat to human life and health in today’s,but it mostly focused on the study of myocardial ischemia injury model in the drug intervention of myocardial ischemia injury model and open form of myocardial ischemia by ligation of myocardial damage model,these models with myocardial ischemia clinical pathogenesis still has very big difference,in addition,the animal model of myocardial ischemia injury standard has yet to be unified and determine,at present,the exercise-induced myocardial ischemia is a important field of sports medicine research,still not mature,but the exercise-induced myocardial ischemia model of exercise-induced myocardial ischemia animal model studies did not see report.Objective:This study through the observation of myocardial cells of rats with HE stained pathological sections,biochemical index,apoptosis index,analysis of rat in different strength,different time of exhaustive degree of myocardial injury in sports,the establishment of exercise-induced myocardial ischemia reperfusion injury in rats model,in order to provide a simple myocardial ischemia injury model of repeatability,but also provide some basis for exercise-induced myocardial ischemia injury.Methods:This experiment to six weeks of age grade clean healthy SD rats as the research object,the random divided into 12 groups,namely,the quiet control group(X),exhaustion exercise high strength once group(A),exhaustion exercise high strength three times group(B),exhaustion exercise high strength a week group(C),exhaustion exercise high strength two weeks group(D),High strength exhaustion exercise group(E),exhaustion exercise medium strength once group(F),exhaustion exercise medium strength three times groups(G),exhaustion exercise medium strength a week group(H),medium strength exhaustion exercise in two weeks(I),exhaustion exercise medium strength three weeks group(J),isopropyl adrenaline(ISO)model of the control group(M),six in each group,group no difference between weight and high strength of 30 M/min,0 degrees,the strength of 20 M/min,0 degrees,exercise to exhaustion of rats,recording time,HE pathological analysis of structure of myocardial injury,colorimetric method and ELISA method to detect the serum myocardial injury,RT-PCR detection of myocardial cell apoptosis.Results:1,Myocardial HE dyeing test results show that with the extension of the damage degree of athletic time more and more serious,and the intensity of high intensity exercise group than medium strength exhaustion exercise group of damage is more serious,high strength exhaustion sports training needs three weeks to reach the damage degree of the ISO model group,the rest of the group take longer to reach the damage degree of the ISO model group;2,Rat serum AST activity:ISO is extremely significant higher than quiet control group,model control group exhaustion exercise group and control group comparison:medium strength in one week,the high strength three group and high strength one week group significantly higher than that of quiet control group,medium strength in the two weeks,medium strength in the three weeks group,high strength two week group,high strength three weeks group of extremely significantly higher than that of quiet control group,the exhaustion sports groups compared with ISO model control group,the strength of a group,the intensity in the three group,high strength,high strength,a group of three groups of AST activity significantly below the ISO model control group,when the movement time to a week,which group is the AST activity and ISO model control group there was no significant difference,and with the extension of time,medium strength in the three weeks group,high strength two week group,high strength three weeks group AST activity more than ISO model control group;3,Rat serum LDH activity:ISO model control group significantly higher than that of quiet control group,exhaustion exercise group and quiet control group comparison:medium strength a week group significantly higher than that of quiet control group.Medium strength two weeks group,medium strength three weeks group,high strength one week group,high strength two weeks group,high strength three weeks group extremely significantly higher than that of quiet control group.The exhaustion sports groups compared with ISO model control group comparison:medium strength once group,high strength once group extremely significantly lower than the ISO model control group.Medium strength three times group,high strength three times group significantly lower than the ISO model control group.And starting from the exhaustion sports week group,each group of LDH activity and ISO model control group,there was no significant difference.Medium strength two weeks group,medium strength three weeks group,high strength two weeks group,high strength three weeks group of LDH activity than the ISO model control group,high strength group than medium strength group is more obvious;4,The rat serum cTn-I activity:ISO model control group significantly higher than that of quiet control group,exhaustion exercise group and quiet control group comparison:high strength week group significantly higher than that of quiet control group,medium strength two weeks group,medium strength three weeks group,high strength two weeks group,high strength three weeks group of extremely significantly higher than that of quiet control group.The exhaustion sports groups compared with ISO model control group:medium strength once group,medium strength two times group,high strength once group,high strength three times group significantly lower than the ISO model control group.And starting from the exhaustion sports week group,each group of LDH activity and ISO model control group,there was no significant difference.Medium strength two weeks group,medium strength three weeks group,high strength two weeks group,high strength three weeks group of LDH activity than ISO model control group,high strength group than medium strength group is more obvious;5,ISO model control group compared with the quiet control group:the Bcl-2 gene expression is extremely significantly lower than the ISO model control group,Bax gene expression is extremely significant higher than the ISO model control group,the Bcl-2/Bax ratio is extremely significantly lower than the quiet control group,the exhaustion exercise group and quiet control group comparison:the Bcl-2 gene expression,medium strength three weeks group extremely significantly lower than the quiet control group,the rest of the campaign group of extremely significantly lower than the quiet control group,the Bax gene expression the campaign group of extremely significantly higher than the quiet control group,the Bcl-2/Bax gene the campaign group of extremely significantly lower than the quiet control group,the exhaustion exercise group ISO model control group compared:the Bcl-2 gene expression in addition to high strength three weeks group showed no significant change,the rest of the sports group is significantly higher than the ISO model control group,Bax gene expression medium strength three weeks group extremely significant lower than the ISO model control group,high strength three weeks group of extremely significantly higher than the ISO model control group,medium strength two weeks group extremely significant lower than the ISO model control group,High strength one week group,high strength two weeks group extremely significantly higher than the ISO model control group.the Bcl-2/Bax ratio high strength three weeks group of extremely significantly lower than the ISO model control group,high strength two weeks group significantly higher than the ISO model control group,the rest of the sports group of extremely significantly higher than the ISO model control group.Conclusion:1,Exercise-induced myocardial ischemia injury model can be used in medium strength,high strength of exhaustion movement is modeled,and the damage degree of high strength group than medium strength,the time required is shorter.With exhaustion exercise medium strength three weeks,high strength exhaustion exercise for two weeks to reach the level of ISO induced myocardial injury in rats,medium strength exhaustion exercise intensity in three weeks,high strength exhaustion exercise two weeks began the motion model can be used as exercise-induced myocardial ischemia injury model.2,HE pathologic analysis,myocardial enzyme spectrum of serum(AST,LDH),cTn-I cell apoptosis regulation genes,etc.Index modeling at the same time,such as high strength exhaustion exercise than medium strength exhaustion exercise in rat myocardial injury,in a shorter period of time can achieve the damage degree of ISO model control group,and myocardial enzyme spectrum detecting serum and cTn-I earlier than HE staining detection can detect myocardial injury.HE pathological analysis,myocardial enzyme spectrum of serum AST,LDH,cTn-I can be used as exercise-induced myocardial ischemia injury model of evaluation index,the cell apoptosis control gene also can be used as the reference,but it is not the most sensitive indicator.
