Background The protein encoded by TSPAN6(Transmembrane4 Superfamily Member 6)is a member of the Transmembrane 4 Superfamily,which is also known as the tetramine protein family.The TSPAN protein family is a family of transmembrane glycoproteins characterized by the structurally present presence of four hydrophobic domains,three intracellular small regions,and two extracellular rings(EC1 and EC2),in which the variable EC2 region contains specific sites to facilitate interactions with partner proteins.TSPAN proteins can establish unique platforms on the cell surface called TSPAN enrichment microdomains(TEMs).In the microdomain of TSPAN protein to match the other family protein,can be combined with integrin,adhesion molecule and growth factor receptor,matrix metalloproteinases and intracellular signaling molecules,and even members of the family of TSPAN,play a regulating function,through the role of the associated with the TEMs,play to regulate cell migration,adhesion,fusion,signal transduction,and even the role of the transfer.Few studies have been done on TSPAN6,and its function and related mechanism in glioblastoma(GBM)remains unclear.This study aims to explore the possible role of glioma in tissue specific expression,prognostic value and signaling pathway enrichment,predict its function in GBM,and explore the possibility of glioma as a tumor therapeutic target through analysis based on cancer Genome Atlas(TCGA)data set.Methods GEPIA2,UALCAN and Oncolnc were used to collect and analyze the transcriptome expression profile and survival prognosis of TSPAN6 in glioma.The similar gene set of TSPAN6 gene was obtained from GEPIA2 analysis,and the KEGG and GO functional enrichment analysis of the gene set was performed by R Studio.GEPIA2 and R Studio were used to analyze the correlation between the transcriptome expression of TSPAN6 gene and the transcriptome expression of genes related to promoting cancer,proliferation and migration.Finally,the clinical data of 703 glioma patients were analyzed by Cox univariate regression analysis of TSPAN6 gene and prognosis factors.Results Transcriptome expression of TSPAN6 was significantly increased in low-grade gliomas(LGG)and glioblastoma(GBM)compared with non-tumor brain tissues(< 0.05);The transcriptome expression of TSPAN6 was significantly correlated with the overall survival rate of glioma patients(< 0.05).The results of functional enrichment analysis of similar gene sets of TSPAN6 showed that TSPAN6 was correlated with cilia,vesicle formation,cell movement and other functions,which may affect the proliferation,migration and invasion of glioma.The results of intermolecular correlation analysis showed that TSPAN6 gene was positively correlated with the transcriptome expression of PCNA gene(P= 0.039.R=0.16)and negatively correlated with the expression level of PTEN gene(P=0.023,R=-0.18).It was positively correlated with the expression levels of Vimentin andα-SMA in mesenchymal transition(<;0.05,R=0.531,0.228).Conclusion The transcriptome expression of TSPAN6 is significantly higher in human glioma than in non-tumor brain tissue,which is correlated with the overall survival rate(OS)of patients,and may play a role in the process of tumor proliferation,invasion and migration.The expression of TSPAN6 may be of certain value in predicting the occurrence,progression and prognosis of brain glioma.
