| ObjectiveThe level of homocysteine(HCY)in the embryonic developmental arrest group and the normal pregnancy group and enzymes related to folic acid metabolism,namely methylenetetrahydrofolate reductase(MTHFR),methionine synthase reductase(MTHFR),and synase reductase,MTRR)gene polymorphism,analyze the correlation between HCY level and embryonic developmental arrest and MYHFR,MTRR gene polymorphism,and then further analyze the correlation between MTHFR,MTRR gene polymorphism and embryonic developmental arrest.To explore the causes of embryonic developmental arrest from the perspective of genes,to provide more reliable laboratory evidence for future pre-pregnancy genetic counseling and clinical guidance on prenatal and postnatal care.MethodsA total of 63 women who were diagnosed with abortion in the gynecology and obstetrics outpatient department and inpatient department of the First Affiliated Hospital of Dali University from July 2020 to October 2021 were selected as the abortion group.63 women with normal pregnancy(without any adverse pregnancy history and complications,this is a normal pregnancy)were included in the normal group.Enzyme cycling method was used to detect HCY levels in the two groups,and high-resolution melting curve method was used to detect the single nucleotide polymorphism(SNP)of MTHFR and MTRR gene loci in the two groups.The correlation between cysteine level and folate metabolism ability related enzyme gene polymorphism and fetal abortion was analyzed.Result1.There was no significant difference in mean age,height and body mass index between the fetal arrest group and the normal group(P=0.67,0.18,0.51>0.05).2.The mean value of HCY in the fetal arrest group was greater than that in the normal group(8.74±3.40>7.15±2.16),and the difference was statistically significant(P=0.002<0.05).3.The mean value of HCY in the MTHFR C677T site was TT>CT>CC(11.11±3.06>7.79±2.92>7.26±2.42),and the difference was statistically significant(F=12.153,P=0.000<0.05).The mean value of HCY in MTHFR A1298C locus was CC0.05).The mean value of HCY in MTRR A66G site was GG>AG>AA(8.87±2.51>8.29±3.17>7.56±2.83),and the difference was not statistically significant(F=1.373,P=0.257>0.05).4.In MTHFR C677T,the frequency of CC type fetal arrest group was lower than that of the normal group(33.3%<57.1%),and the difference was statistically significant(x~2=12.14,P=0.00<0.05);the frequency of CT type fetal arrest group was higher than that of the normal group(47.6%>38.1%),the difference was statistically significant(x~2=4.41,P=0.04<0.05);the frequency of TT type fetal arrest was significantly higher than that of the normal group(19.0%>4.8%),but the difference was not statistically significant(x~2=0.19,P=0.67>0.05);the T allele frequency in the fetal arrest group was significantly higher than that in the normal group(42.9%>23.8%),and the difference was statistically significant(x~2=10.286,P=0.001<0.05).MTHFR gene C677T locus CT type and TT type carriers had 2.580 times and 6.878 times the risk of fetal abortion,respectively,and the difference was statistically significant(P=0.025,0.010<0.05);5.In MTHFR A1298C,the frequency of AC type in the fetal arrest group was lower than that in the normal group(28.6%<30.2%),and the difference was statistically significant(x~2=4.42,P=0.04<0.05);the frequency of C allele in the fetal arrest group was higher than that in the normal group group(22.2%>15.1%),the difference was not statistically significant(x~2=2.12,P=0.15>0.05).6.The frequency of G allele in MTRR A66G was significantly higher than that in the normal group(34.9%>19.8%),and the difference was statistically significant(x~2=7.21,P=0.007<0.05).MTRR gene A66G locus carriers of AG type had a 2.679-fold higher risk of fetal abortion than AA type,and the difference was statistically significant(P=0.028<0.05).Conclusions1.There is no correlation between the age,height and BMI of the research subjects and the occurrence of fetal abortion.2.The change of HCY level is related to the occurrence of fetal abortion,and the increase of HCY level may increase the risk of fetal abortion.3.The polymorphism of MTHFR C677T gene is related to the increase of plasma HCY level in pregnant women,and the TT genotype may be related to high level of HCY.The gene polymorphisms of MTHFR A1298C and MTRR A66G were not associated with elevated plasma HCY levels in pregnant women.4.The MTHFR C677T gene polymorphism is associated with the occurrence of fetal abortion,and the increase of the frequency of heterozygous mutant genotype(CT)and T allele frequency is related to the occurrence of fetal abortion.The homozygous mutant genotype(TT)increased frequency may be associated with the occurrence of fetal abortion.Homozygous mutant genotype(TT)and heterozygous mutant genotype(CT)are risk factors for fetal abortion.5.MTHFR A1298C gene polymorphism is not associated with fetal abortion.Neither the homozygous mutant genotype(CC)nor the increased frequency of the heterozygous mutant genotype(AC)were associated with fetal arrest.Increased C allele frequency was not associated with fetal arrest.6.The increase in the frequency of MTRR A66G homozygous mutant genotype(GG)has no relationship with the occurrence of fetal abortion.Heterozygous mutant genotype(AG)is a risk factor for fetal abortion.An increase in the frequency of the G allele is associated with the occurrence of fetal arrest. |
