【 Objective 】 To determine the expression of CTHRC1 proteins in extrahepatic cholangiocarcinoma and adjacent tissues.The purpose of this study is to determine the incidence and progression of extrahepatic cholangiocarcinoma.Then,to establish a link between CTHRC1 and various pathological subtypes of patients.Finally,we examine whether CTHRC1 can participate in the epithelial-mesenchymal transition(EMT)process and promote extrahepatic cholangiocarcinoma invasion and metastasis when combined with biomarker of EMT.Additionally,the study aims to provide patients with evidence-based medicine.【Materials and Methods】1.Twenty-five specimens of extrahepatic cholangiocarcinoma tissues and twenty-five specimens of adjacent tissues(>2cm away from the tumor edge)were collected at Dali University’s first affiliated hospital between January 2018 and December 2021.The pathological and clinical data on the patients were complete.During this period,fifteen specimens of fresh cholangiocarcinoma tissue were collected,as well as fifteen specimens of adjacent tissues following radical surgery.Extrahepatic cholangiocarcinoma tissues were used in the experimental group,whi-le normal liver tissues were used in the control group.2.CTHRC1 expression is detected using immunohistochemistry and western blot in extrahepatic cholangiocarcinoma tissues and adjacent tissues.The purpose of this study is to determine the role of CTHRC1 in the occurrence and progression of extrahepatic cholangiocarcinoma,as well as the relationship between various pathological features(age,gender,TNM stage).3.Immunohistochemistry was used to detect the expression of E-cadherin,4.N-cadherin,and MMP9 proteins in extrahepatic cholangiocarcinoma and adjacent tissues.Spearman analysis was used to determine the relationship bet ween the pr-oteins.【Result】1.IHC analysis revealed that CTHRC1 expression was positive in 60%(15/25),of extrahepatic cholangiocarcinoma tissue and 20%(5/25)of adjacent tissues.When combined with the WB result,the two findings indicated that it is significantly greater in extrahepatic cholangiocarcinoma than in adjacent tissues.2.CTHRC1 protein expression is significantly higher in extrahepatic cholangiocarcinoma tumor tissues with lymph node metastasis than in tumor tissues wit-hout lymph node metastasis(P=0.009).It is significantly higher in the nerve invasion group(P=0.034)and is also associated with tumor stage(P=0.022).There is no correlation between age,gender,vascular infiltration,or tumor differentiation of the degree in patients(P>0.05).3.In extrahepatic cholangiocarcinoma tissues,E-cadherin,N-cadherin and MMP9 protein are positive expression rates of 40%,56% and 52%.E-cadherin is significantly higher expression in adjacent tissues than cancer tissues(P<0.05).Ncadherin expression has no significance between cancer tissues and adjacent tissues(P>0.05).MMP9 is significantly higher expression in adjacent tissues than cancer tissues(P<0.05).4.In extrahepatic cholangiocarcinoma tissues,the negative correletion between the expression of CTHRC1 and E-cadherin(rs=-0.500,P=0.011).The positive correlation between the expression of CTHRC1 and N-cadherin(rs=0.428,P=0.033).The positive correlation between the expression of CTHRC1 and MMP9(rs=0.400,P=0.048).The negative correlation between the expression of E-cadherin and Ncadherin(rs=-0.592,P=0.002).The negative correlation between the expr-ession of Ecadherin and MMP9(rs=-0.582,P=0.002).The positive correlation bet-ween the expression of N-cadherin and MMP9(rs=0.345,P=0.092).【Conclusion】1.The positive expression of CTHRC1 protein in extrahepatic cholangiocarcinoma,CTHRC1 may be involved the occurrence of extrahepatic cholangiocarcin-oma process.2.In extrahepatic cholangiocarcinoma tissues,the expression of CTHRC1 is significantly related with lymph node metastasis,nerve infiltration and TNM stage.And it may become one of the molecular biomarker to estimate the prognosis of the patients.3.CTHRC1 may be promote the invasion and metastasis of extrahepatic cholangiocarcinoma by the process of EMT.
