Objective To investigate the relationship between the expression of CO-029 and epithelial mesenchymal transition(EMT) induced by tumor necrosis factor alpha(TNFα) in intrahepatic cholangiocarcinoma cell. Methods RT-PCR and Western blot were used to detect the Co-029 expression in ICC cell and tumor tissues; lentivirus interfering CO-029 expression was included to investigate its effect on EMT process induced by TNFα. Western blot and mass spectrometry after immunoprecipitation were used to confirm that the TNFαR1 can be formed into the complexes directly or indirectly with the transmembrane protein CO-029 in intrahepatic cholangiocarcinoma. Results There are different degrees expression of CO-029 in intrahepatic cholangiocarcinoma cells and tumor tissues, lentiviral interference significantly reduced expression of CO-029 in intrahepatic cholangiocarcinoma cells. TNFα failed to induce EMT process when the CO-029 expression was decreased. TNFαR1 in intrahepatic cholangiocarcinoma could directly or indirectly form complexes with transmembrane protein CO-029. Conclusion CO-029 could mediate TNFα/TNFαR1 signaling to induce EMT in cholangiocarcinoma cells, promoting the ability of invasion and metastasis in intrahepatic cholangiocarcinoma.
