Ginger is the fresh rhizome of Zingiber officinale Rosc.,a common Chinese medicine,which is mostly used for cold and cold,stomach cold vomiting,cold phlegm cough and fish and crab poisoning.It is also a traditional condiment.6-gingerol is the main active component of ginger.Modern pharmacological studies have proved that 6-gingerol has many biological activities,such as anti-inflammatory,liver protection,cholagogue and anti-tumor.It is also found that 6-gingerol has protective effect on non-alcohol induced liver injury in mice.Tongling white ginger is a rare and excellent local variety of ginger,one of "eight treasures of Tongling",which has a long cultivation history and is rich in nutrients.In this study,firstly,whether 6-gingerol can protect mice from alcoholic liver injury was discussed.Based on the protective effect of 6-gingerol on mice with alcoholic liver injury,the protective effect of Tongling white ginger on mice with alcoholic liver injury was further studied.The following experiments were carried out in this subject:1.Protective effect of 6-gingerol on alcoholic liver injury in mice(1)In this experiment,the model of alcoholic liver injury in mice was established by Gao Bin model method,and the mice were treated by gavage.The animals were divided into the following groups: normal group(Vehicle),alcohol group(Et OH),6-gingerol high-dose group(100 mg/kg),6-gingerol middle-dose group(50 mg/kg),6-gingerol low-dose group(25 mg/kg)and Silymarin group(100 mg/kg).The liver injury and the expression level of pro-inflammatory factors were detected by liver histopathology(HE staining,oil red O staining),serology(AST,ALT,T-CHO,TG content determination)and ELISA.The expression level of TLR4 in mouse liver was detected by immunohistochemistry,and the expression levels of TLR4 and inflammatory factors were detected by protein immunoblot.(2)The damage of small intestine tissue in mice with alcoholic liver injury was detected by intestinal histopathology(HE staining),the expression level of TLR4 in small intestine was detected by immunohistochemistry,and TLR4 and tight junction protein(zonula occludes-1;ZO-1),Occlaudin and claudin-1,and 16 S r RNA sequencing method was used to detect the taxonomic changes of microbial communities in mouse feces.2.The protective effect of Tongling white ginger on alcoholic liver injury in miceIn this experiment,the model of alcoholic liver injury in mice was established by Gao Bin model method,and the mice were treated by gavage.The animals were divided into the following groups: normal group(Vehicle),alcohol group(Et OH),Tongling white ginger(GN)low dose group(0.5 g/kg),Tongling white ginger(GN)middle dose group(1g/kg),Tongling white ginger(GN)high dose group(1.5 g/kg)and Silymarin group(Silymarin,0.1 g/kg).The liver injury and the expression level of pro-inflammatory factors were detected by liver histopathology(HE staining,oil red O staining),serology(AST,ALT,T-CHO,TG content determination)and ELISA.Results:1.(1)6-gingerol has protective effects on alcoholic liver injury: HE staining,oil red O staining,AST,ALT,T-CHO,TG content determination,suggesting that 6-gingerol can significantly improve the liver injury caused by alcohol,and the results of ELISA showed that 6-gingerol can significantly reduce the high expression of IL-1β,IL-6 and TNF-αcaused by alcohol.Immunohistochemistry showed that 6-gingerol could reduce the high expression of TLR4 induced by alcohol.The results of protein Western blot showed that6-gingerol could significantly improve the abnormal expression of TLR4 and inflammatory factors caused by alcohol.(2)Intestinal histopathology(HE staining),immunohistochemistry and protein Western blot showed that 6-gingerol could improve the intestinal tissue damage caused by alcohol in mice,and 16 S r RNA sequencing showed that 6-gingerol obviously reversed the changes of microbial taxonomy in feces caused by alcohol.2.Tongling white ginger has protective effects on alcoholic liver injury: HE staining,oil red O staining,AST,ALT,T-CHO,TG content determination,suggesting that Tongling white ginger can significantly improve the liver injury caused by alcohol,and the results of ELISA test indicate that Tongling white ginger can significantly reduce the high expression of IL-1β,IL-6 and TNF-α caused by alcohol.Conclusion:1.6-gingerol has a protective effect on alcoholic liver injury in mice.The protective effect of 6-gingerol may be to inhibit the transduction of inflammatory signal pathway by down-regulating the expression of TLR4 protein,thus effectively alleviating alcohol-induced liver injury.6-gingerol can inhibit the secondary impact of enterogenous factors on the liver by reducing the changes of intestinal microecology caused by alcohol,and indirectly reduce the liver damage caused by alcohol.2.Tongling white ginger has protective effect on mice with alcoholic liver injury.
