Selenium-enriched Yeast Combined With Vitamin E Attenuates Cadmium-induced Liver Injury In Mice

Objective: By observing the protective effect of selenium enriched yeast combined with vitamin E on chronic liver injury induced by cadmium chloride in mice,new clinical solutions and research ideas are provided for the development of treatment of liver injury caused by heavy metals.Methods: In this experiment,50 SPF Kunming mice of appropriate age were randomly divided into negative control group,cadmium chloride group(5mg/kg),selenium-enriched yeast group(100mg/kg),vitamin E group(50mg/kg)and selenium-enriched group Yeast(100mg/kg)+Vitamin E group(50mg/kg),The corresponding reagent was given once a day in the afternoon for 28 consecutive days.The serum alanine aminotransferase(ALT),aspartate aminotransferase(AST)and lipid peroxidation product malondialdehyde(MDA)were determined by the Lai’s method using related kits,and observed under a light microscope.The pathological sections of liver tissue were used to semi-quantitatively detect and analyze the expression levels of related proteins in liver tissue by Western blotting.Results: By measuring serum transaminase and observing the results of HE staining,it can be concluded that cadmium chloride can cause liver injury,and the combined drug group can reduce this injury;Selenium enriched yeast combined with vitamin E can increase the expression of antioxidant proteins cat,GPx4,nuclear Nrf2 and SOD1,down regulate the expression of liver apoptotic proteins Bax,cleaved caspase-9 and cleaved Caspase-3,up regulate the expression of Bcl-2 and down regulate TNF-α And NF-k B p65 expression,so as to improve the degree of liver injury;Selenium enriched yeast combined with vitamin E may reduce liver injury caused by cadmium exposure by activating PI3K/Akt signaling pathway.Conclusion: Selenium-enriched yeast combined with vitamin E can alleviate cadmiuminduced liver damage.
