Objective To explore the active components and mechanism of Huang Long Zhike Granule in the treatment of Cough Variant Asthma by network pharmacology.Methods Firstly,the components of Huang Long Zhike Granule were searched through the Chinese Medicine System Pharmacology(TCMSP)database and the articles from CNKI and Pubmed databases.The targets of the active components were predicted by TCMSP database.Cytoscape 3.8.2 was used to build a network between active components and targets.Cough Variant Asthma disease targets were obtained in Genecards database and Disgenet database,and the venn diagram was drawn to obtain overlapping targets.Use the String platform to construct a Protein-protein interaction network of overlapping targets.then mapped with drug targets to obtain the predicted targets of Huang Long Zhike Granule for Cough Variant Asthma disease.The target interaction network was imported into Cytoscape 3.8.2 to screen the core targets of Huang Long Zhike Granule.Finally,Metascape database were used to perform GO biological process enrichment analysis and KEGG pathway enrichment analysis.Results The results of our network pharmacology research showed that the Huang Long Zhik e Granule,composed of 5 Chinese herbs,contained 73 main components,and 178 genes rela ted to Cough Variant Asthma.The key active ingredients include quercetin,luteolin,kaempfe rol,wogonin,yohimbine,beta-sitosterol,etc.The core targets include: EGFR,PTGS2,JUN,M APK3,MAPK1,MAPK8,MMP9,IL-1β,EGF,MYC,IL-6,CCND1,TNF,TP53,VEGFA,ESR1,CASP3,FOS,CXCL8,AKT1.GO biological process analysis obtained 687 biological process es,mainly involved in cellular response to cadmium ion,cellular response to reactive oxygen species,cellular response to oxidative stress,response to lipopolysaccharide,response to mo lecule of bacterial origin,response to inorganic substance and positive regulation of cell migr ation,etc.Cellular component analysis obtained 18 cellular components,mainly involved in membrane raft,membrane microdomain,membrane region,vesicle lumen,RNA polymerase II transcription factor complex,nuclear transcription factor complex,nuclear envelope,adhe rens junction,anchoring junction,late endosome,etc.Molecular Function analysis obtained molecular function,mainly involved in cytokine receptor binding,nitric-oxide synthase regul ator activity,transcription factor binding,phosphatase binding,MAP kinase activity,cytokin e activity,receptor ligand activity,receptor regulator activity,protein kinase activity,growth factor receptor binding,etc.KEGG signaling pathway analysis obtained 138 signaling pathw ays,mainly related to: MAPK signaling pathway,IL-17 signaling pathway,TNF signaling pa thway,AGE-RAGE signaling pathway in diabetic complications,PI3K-Akt signaling pathwa y and Toll-like receptor signaling pathway,etc.Conclusion This study preliminarily revealed the pharmacodynamic substance basis of Huang Long Zhike Granule and its possible mechanisms for treating Cough Variant Asthma,indicating that Cough Variant Asthma can be treated by Huang Long Zhike Granule through“mul-target and multi-pathway”.providing reference for the study of Huang Long Zhike Granule in the treatment of Cough Variant Asthma.
