| ObjectiveThrough the establishment of a gastric precancerous lesion(GPL)rat experimental model,the effects of Weiweixiao on gastric mucosal lactic acid content,MCT1,MCT4and CD147 protein expression and m RNA levels in GPL rats were observed,aiming to reveal that Weiweixiao improves the acidic microenvironment,to intervene or block the effect mechanism of gastric precancerous lesions in rats.MethodsSeventy-four SD male rats were randomly divided into 12 normal groups and 62models were made.The GPL rat model was induced and replicated by the MNNG-ammonia complex method for the model building.At the 9th week,the model rats were randomly divided into the model group,the folic acid group(2.7 mg·kg-1),Low-dose group(12.6,6.3,3.15 g·kg-1),12 rats in each group,gavage treatment lasted for 12weeks,observe the general condition of the rats.After the experiment,two pieces of mucosal tissue at the junction of the gastric body and antrum of the rat were taken,and the histopathological changes of the gastric mucosa were observed by HE staining,the lactic acid content of the gastric mucosal tissue was detected by chemical colorimetry,and the stomach was detected by immunohistochemistry and real-time fluorescence quantitative PCR.Mucosal tissue MCT1,MCT4,CD147 protein expression and m RNA level.Results1.Effects on the general condition of ratsRats in the model group were sluggish,with yellowish and dull fur,severe hair loss,malnutrition,significantly lower body weight,reduced water and food intake,and loose stools;after the treatment of stomach atrophy,the general condition of each dose group was normal.There are different degrees of improvement.2.Effect on HE staining of gastric mucosal tissueThe gastric mucosal epithelial gland structure and arrangement of rats in the model group are disordered,the boundary is not clear,the number of glands is reduced,the number of glands is reduced,and the stratification and back-to-back changes can be seen,there are deep staining and enlargement of nuclei,increased nucleoplasm ratio,and significantly weakened nuclear polarity,It can be seen that mitosis increased;after Weiweixiao treatment,the pathological manifestations of the gastric mucosal epithelial gland structure,arrangement disorder and cell atypia of rats in each dose group were improved.3.Effect on the content of lactic acid in gastric mucosaThe gastric mucosal lactic acid content of rats in the model group was significantly higher than that of the normal group(P<0.01);after Weiweixiao treatment,the gastric mucosal lactic acid content of rats in each dose group was significantly reduced(P<0.05,P<0.01).4.Influence on the expression level of MCT1 protein and m RNAThe expression levels of MCT1 protein and m RNA in the model group were sig-nificantly higher than those in the normal group(P<0.05,P<0.01);after Weiweixiao treatment,the expression levels of MCT1 protein and m RNA in each dose group de-creased,but the difference was not statistically significant(P>0.05).5.Effect on MCT4 protein and m RNA expression levelThe expression level of MCT4 protein and m RNA in the model group was signif-icantly higher than that in the normal group(P<0.01);after Weiweixiao treatment,the expression of MCT4 protein in each dose group was significantly reduced(P<0.01),and the level of MCT4 m RNA in the high and medium dose groups was significant Decrease(P<0.05,P<0.01).6.Influence on the expression level of CD147 protein and m RNAThe expression of CD147 protein and m RNA in the model group was significantly higher than that in the normal group(P<0.01);after Weiweixiao treatment,the expres-sion of CD147 protein in each dose group was significantly reduced(P<0.05,P<0.01),and the CD147 m RNA in the high-dose group The level was significantly reduced(P<0.05).Conclusion1.Weiweixiao can effectively improve the general conditions and histopathologi-cal changes of gastric mucosa in GPL rats,and curb the malignant transformation of gastric mucosal epithelium.2.The content of lactic acid in the gastric mucosa of GPL rats increases.Wei-weixiao can reduce the content of lactic acid in the gastric mucosa of GPL rats and correct the acidic microenvironment of the gastric mucosa.3.MCT1,MCT4 and CD147 are overexpressed in gastric mucosal tissues of GPL rats.The key regulatory molecules of Weiweixiao down-regulating the acidic microen-vironment may be MCT4 and CD147,but Weiweixiao has no intervention effect on MCT1. |
