Objective To study the expression of TLR4 protein in colorectal cancer tissues and adjacent tissues,and to analyze the correlation between TLR4 protein,preoperative serum CEA levels and clinicopathological data of colorectal cancer patients,and analyze colorectal cancer patients.Correlation between preoperative serum CEA level and TLR4 protein expression.Methods(1)Retrospective collection of colorectal cancer tissue resectioned by General Surgery from the Gezhouba Group Central Hospital(the Third Clinical College of China Three Gorges University)from August 2015 to August 2017,and after embedding in our hospital pathology department The paraffin blocks were confirmed by pathological examination as colorectal adenocarcinoma after surgery.A total of 45 cases were taken as the experimental group.All patients with colorectal cancer received no radiotherapy,chemotherapy,and hormone immunotherapy before surgery.At the same time,the control group consisted of 39 cases of paracancerous tissue.The 45 cases of colorectal cancer in the above specimens of the distant colon tissue of the lesion(6cm outside the margin of the cancer,and pathologically confirmed no tumor cells)of the intestinal mucosa tissue as a control group.Immunohistochemical staining was used to detect the expression of TLR4 in 45 cases of colorectal cancer tissues and 39 cases of adjacent tissues.(2)To collect and sort out the clinicopathological data of 45 patients with colorectal cancer,and use the Fisher exact test to analyze the correlation between TLR4 protein expression and clinicopathological data using SPSS19.0 software.(3)Collection of preoperative serum CEA results in 45 patients with experimental group(cancer tissue).Judgment criteria:0-5ng/ml negative,>5ng/ml positive,using SPSS19.0 software,Fisher’s exact probability method was used to analyze the correlation between preoperative serum carcinoembryonic antigen level and clinicopathological data of patients with colorectal cancer.(4)The correlation between TLR4 protein expression and preoperative serum CEA in colorectal cancer patients was analyzed by SPSS19.0 software,correlation analysis and Kappa test.Results(1)TLR4 has different levels of expression in colorectal cancer tissues,and the positive expression rate in colorectal cancer tissues is 68.9%(31/45),and the positive expression rate in the adjacent tissues is 10.3%(4/39).The difference of positive expression rate between the two groups was statistically significant(P<0.05).Dyeing,TLR4 protein expression in colorectal cancer tissue samples of cytoplasm,especially in poorly differentiated colorectal cancer tissues,dark brown,times,and differentiation of colon cancer tissues in high differentiation,the pale brown,but it is still a corresponding tissue adjacent to carcinoma.(2)colorectal cancer tissue pathology in high differentiation group of TLR4 protein positive rate was 23.1%(3/13),in the positive expression rate of low differentiation group was 87.5%(28/32),the positive expression rate of difference between two groups was statistically significant(P<0.05);The positive expression rate of TLR4 protein in the whole group was 90.0%(27/30),and the positive expression rate of the subserous group was 26.7%(4/15),and the difference in the positive expression rate between the two groups was statistically significant(P<0.05).With lymph node metastasis group of TLR4 protein positive expression rate is 89.3%(25/28),no positive expression rate of lymph node metastasis was 35.3%(6/17),the positive expression rate of difference between two groups was statistically significant(P<0.05);The positive expression rate of TLR4 protein in group A+B in Duke’s stage was 42.1%(8/19),and the positive expression rate of C+D group was 88.5%(23/26),and the difference of positive expression rate between the two groups was statistically significant(P<0.05).In the two groups and gender groups,there was no statistically significant difference between the two groups of TLR4 protein expression(P>0.05).(3)the positive rate of serum CEA was 38.5%(5/13)in the high-differentiation group and 68.8%(22/32)in the middle and low differentiation group.There was no significant difference in the positive expression rate between the two groups(P>0.05).With lymph node metastasis of preoperative serum CEA positive rate was 75.0%(21/28),with no lymph node metastasis positive rate was 35.3%(6/7),the positive expression rate of difference between two groups was statistically significant(P<0.05);The positive rate of serum CEA was 60.0%(18/30)and 60.0%(9/15)in the subserous infiltration group,and there was no significant difference in the positive expression rate between the two groups(P>0.05).The positive rate of CEA was 31.6%(6/19)and 80.1%(21/26)in the group C+D group,and the difference in positive expression rate between the two groups was statistically significant(P<0.05).Preoperative serum CEA level was expressed in the age and gender of colorectal cancer patients,and there was no statistically significant difference in the positive rate between the two groups(P>0.05).(4)the positive expression rate of TLR4 protein in colorectal tissues was 68.9%(31/45),preoperative serum CEA positive rate was 60.0%(27/45),TLR4 protein in colorectal cancer tissues and preoperative serum CEA positive rate 46.6%(27/58)at the same time,both the negative rate was 18.8%(6/32).Using the correlation analysis of 2-by-2 paired data,it was found that the Pearson contingency table coefficient r=0.445(P<0.05),which was statistically significant.At the same time,the Kappa value was 0.449(P=0.001),which was statistically significant.Conclusion Conclusion:(1)the TLR4 protein exists in colorectal cancer tissues and had a higher expression,and LTR4 protein expression in colorectal cancer tissue samples of cytoplasm,especially in poorly differentiated colorectal colorectal tissues,dark brown,second,differentiated colorectal carcinoma tissues in high differentiation,the pale brown,but still a tissue adjacent to carcinoma apparently normal mucosa.(2)the expression of TLR4 protein may be associated with colorectal cancer in certain pathological biological characteristics,the lower the degree of tumor differentiation,the higher the Duke ’s stage,the deeper the infiltration degree and lymph node metastasis,the higher the expression of TLR4,hint it may participate in taking colorectal cancer progression.(3)preoperative serum CEA level of patients with colorectal cancer was correlated with tumor metastasis and Duke’s stage,suggesting that it was related to prognosis of patients with colorectal cancer.(4)the expression of TLR4 protein in colorectal cancer has certain correlation with preoperative serum CEA level,and both have certain consistency,potential to provide more clinical laboratory diagnosis of colorectal cancer.
