ObjectiveAcupuncture with acupoints is based on the theoretical system of traditional Chinese medicine,combined with the rules of the meridian and the functions and characteristics of the acupoints,and then two or more acupoints are used in the same way through traditional Chinese medicine dialectics,and acupuncture or moxibustion is used to achieve the purpose of treating diseases.In this study,a rat model of insomnia was selected,and different acupoint compatibility methods were used for acupuncture treatment.The changes in the content of Melatonin(MT)in the conarium of rats were observed and compared,and different acupoint compatibility methods were used to treat insomnia.Effect.Methods60 normal SD rats were randomly assigned to 5 groups: blank group,model group,Treatment group 1(Baihui,Shenmen),treatment group 2(Baihui,Sanyinjiao),treatment group 3(Baihui,non-acupoints),12 rats/group,the blank group was given conventional feeding treatment,and the remaining4 groups were restricted The sex fixation method lasted for 15 days.The insomnia model was established by intraperitoneal injection of P-chlorophenylalanine(PCPA)suspension.At the beginning of the experiment,first let each group of rats move freely for 24 hours and record the basis level.For the following 5 days,the three groups of rats in treatment group 1,treatment group 2,and treatment group 3 were treated with corresponding acupuncture points every day.After the treatment,each group of rats underwent a 24-hour spontaneous activity test.After sampling,high performance liquid chromatography(HPLC)and real-time fluorescent quantitative PCR were used to detect the levels of MT and CNMT1 and MT2 m RNA in peripheral blood,respectively.Result1.Establishment of insomnia modelThis study used pentobarbital sodium sleep test.The sleep latency and sleep time of rats in the blank group were not significantly different from the basic level(P>0.05).Compared with before modeling,the sleep latency of rats in the model group,treatment group 1,treatment group 2,and treatment group3 were all higher than before the model was established;the sleep latency of the model group was longer than that of the blank group(P < 0.01),and the sleep time was shorter than that of the blank group(P <0.01),indicating that the model was successful.2.The effect of acupuncture with different acupoints on the activity of ratsAfter giving different acupoints and acupuncture treatments,the 24-hour spontaneous activity level of the rats in each group was tested.The results showed that compared with the blank group,the rats in the model group had significantly increased daytime activity(P <0.01)and nocturnal activity.Decrease(P <0.01).Compared with the model group,the daily activity of treatment group 1,treatment group 2,and treatment group 3 were significantly reduced(P <0.01)and night activity was significantly increased(P <0.01);compared with treatment group 3,treatment The amount of daily activity in group 1 and treatment group 2 decreased(P <0.05),and the amount of night activity increased(P <0.05),but there was no statistical difference between treatment group 1 and treatment group 2(P> 0.05).The activity level of the rats during the rest period was measured.The results showed that after acupuncture treatment with different acupoints,the spontaneous activities of the rats in the treatment group 1,treatment group 2,and treatment group 3 showed a circadian rhythm resynchronized with the light and dark cycle within 24 hours.Compared with the model group,the rest period activity of the rats in the three acupuncture treatment groups was significantly reduced(P <0.05);compared with the treatment group 3,the rest period activity of the treatment group 1 and treatment group 2 decreased,and the difference was statistically significant Scientific significance(P <0.05);there was no statistical difference between the direct results of treatment group1 and treatment group 2.3.The effect of different acupoints and acupuncture on the level of MT m RNA in peripheral blood MT and pineal gland in rats(1)Studies have shown that acupuncture treatment can increase the content of MT protein in the peripheral blood of rats: HPLC test results show that the MT content of the pineal gland in the acupuncture treatment group,treatment group 1,treatment group 2,and treatment group 3 are significantly higher than that of the model Group(P <0.01).Both treatment group 1 and treatment group 2 were higher than treatment group 3(P <0.05),and there was no significant difference between these two groups(P> 0.05).(2)Acupuncture treatment with different acupoints can increase the expression levels of SCN MT1 and MT2 m RNA in rats.The QPCR test results showed that the expression levels of MT1 and MT2 m RNA in the model group were significantly reduced compared with the blank group(P <0.01);Compared with the treatment group,the expression of MT1 and MT2 m RNA in treatment group 1,treatment group 2 and treatment group 3 increased significantly(P <0.01);the expression of MT1 and MT2 RNA in treatment group 1 and treatment group 2 was higher than that in treatment group 3(P<0.05);There was no statistically significant difference in the expression levels of MT1 and MT2 m RNA between treatment group 1 and treatment group 2(P>0.05),but treatment group 1 tended to be higher than treatment group 2.ConclusionAcupuncture can effectively improve the sleep disorder of insomnia model mice and reduce the degree of anxiety.By increasing the content of MT in peripheral plasma and increasing the expression levels of MT1 and MT2 m RNA in the pineal gland,it can treat insomnia,and it is compatible with different acupoints.The method has different acupuncture treatment effects,which proves that the compatibility of acupoints is an important factor influencing the acupuncture treatment effect.The selection of acupuncture points along the meridian can improve the curative effect of acupuncture treatment to a certain extent.
