Objective:To observe the effects of the acupoints of different meridians on the Melatonin?MT?content of the pineal gland and the expression of Melatonin receptor1?MT1?and Melatonin receptor 2?MT2?in the rats,and to explore the effect of the selected acupoints on the distribution of acupoints.Methods:60 SD rats were randomly divided into normal control,model,same meridian:“Bahui”?GV 20?-“Shenmen”?HT 07,3-acupoints?,related meridian:“Bahui”?GV 20?-“Sanyinjiao”?SP 06,3-acupoints?,non-acupoint:“Bahui”?GV 20?-non-acupoint?3-acupoints?groups,with 12 rats in each group.Insomnia model was eatablished by Intraperitoneal injection of para chlorophenylala-nine suspension.Acupuncture therapy once daily for 7 days.Observation rat circadian activity,sleep time,sleep latency,using immune histochemical method to detect the pineal gland of rats MT of grey value,using real-time fluorescent quantitative PCR method to detect the suprachiasmatic nucleus in rats MT1 and MT2 mRNA expression.Results:1?After modeling,pentobarbital righting the experimental results show that the model group the sleep latency than before modeling extension?P<0.05?,sleep duration was shortened before modeling?P<0.05?,and to compare the difference with the control group?P<0.05?,suggesting that the model was successful.Compared with normal group,the sleep latency period of sodium pentobarbital sleep was increased in the model group?P<0.05?,and that of sleep duration.Compared with model group,the sleep latency period was decreased in the same meridian group,related meridian group,non-acupoint group?P<0.05?,and the sleep duration was increased in the same meridian group?P<0.05?.2?Diet:compared with the blank group,the model group to reduce consumption,the difference was statistically significant?P<0.05?;compared with the model group,Baihui plus Shenmen group increase food intake?P<0.05?;comparison between treatment groups,Baihui plus Shenmen group than Baihui and Sanyinjiao group,Baihui and non acupoint group?P<0.05?.The amount of drinking water:model group was less than that of control group?P<0.05?;Baihui plus Shenmen group,Baihui and Sanyinjiao group,Baihui and non acupoint group less than the model group?P<0.05?;the treatment group were no difference?P>0.05?.3?The body weight increase of the rats compared with the control group,model group,body weight growth quantity is very few,the difference was statistically significant?P<0.05?;compared with the model group,Baihui plus Shenmen group,Baihui and Sanyinjiao group weight increase more?P<0.05?;comparison between treatment groups,Baihui plus Shenmen group than Baihui and Sanyinjiao Baihui plus group,non acupoint group?P<0.05?.4?Comparison of the gray value of pineal body:compared with the blank group,MT gray value of the model group was increased;Compared with the model group,the MT gray value of Baihui plus Shenmen group,Baihui plus Sanyinjiao group was significantly reduced?P<0.05?.There was no significant difference in MT gray value between the three groups in each treatment group?P?0.05?,but there was a tendency in the Baihui plus Shenmen group to be lower than the other two groups.5?The mRNA content of MT1 and MT2 was compared between the two groups:compared with the blank group,the mRNA expression of MT1 and MT2 in the model group was significantly reduced?P<0.01?.After acupuncture,compared with the model group,the expression level of MT1 and MT2 mRNA in the Baihui plus Shenmen group,Baihui plus Sanyinjiao group and Bahui plus non-acupoint group was significantly increased?P<0.01?.The expression of MT1 mRNA was significantly higher than that of Bahui plus non-acupoint group?P<0.01?,and the mRNA expression level of MT2 was significantly higher than that of the other two groups?P<0.01?.Conclusion:Acupuncture treatment can increase the MT content of conarium and the expression of MT1 and MT2 mRNA in the suprachiasmatic nucleus of insomnia rats.Fixed local acupoints combined with circulation point selection can improve the compatibility of acupoints.
